Astragaloside IV (AsIV) has the active on treating on viral myocarditis which is the main component of astragaloside extract. AsIV can protect the damaged heart cells, relieving the symptoms other purposes by protecting vascular endothelial by increasing the resistance to oxygen, thus reducing the damage caused by lack of oxygen. AsIV also has the ability of hepatoprotective, anti-aging and many other pharmacological effects. There are still much similar to the saponins AsIV left by macroporous resin adsorption, butanol extraction method, which only strains the extract of Astragalus saponin with AsIV,and that will result in waste of resources and environmental pollution, therefore it is necessary to develop a similar structure through the transformation of the increased astragaloside astragaloside content of new methods. Some components can be obtained through hydrolysis in theory, so it is a viable starting point by the biological synthesis doing it.Biosynthesis of Radix is an effective method to improve the content of astragaloside, but it has to encounter some problems, such as conversion of poor specificity, conversion efficiency obtained AsIV relatively low, it is difficult to achieve the purpose of industrial production. To improve products through the content of biosynthesis, bacteria are the key. In existing reports, only one set of studies is reported that Absida sp. A3r glucosidase can improve AsIV content. So, we consider that the approach in microbial transformation, using mutant strains of transformation method to overcome the difficulties.In this study,we used the originating branch Absidia strains, mutagenesis by UV,DES,NTG, to improve screening efficiency astragaloside content of Radix 31.1% species.After optimization of medium components and optimization of culture conditions, shake flask test results show that the substrate feeding amount is 15 mg / mL, the maximum conversion rate can reach 43.65%.We extracted and purified the products, got high purity monomer. We have got the establishment of a laboratory-scale purified Astragaloside effective method by 1H, 13C-NMR and HPLC data on the product structure to confirm.Such work laid the foundation for industrial production of ASIV. |
