Inhibitory Effect Of Coix Seed Extracting Solution On Rat Corneal Neovascularization

Objective: Normal corneal is transparent and avascular, transparent corneal takes an important part in visual function. Infection,trauma, immune reaction, such as pathological conditions can lead to the formation of corneal neovascularization(CNV), The formation of CNV due to various factors, VEGF is the most important promote angiogenesis factor in present. Coix Seed is traditional Chinese medicine by containing various bioactive substances, in recent years, Chinese and foreign scholars has reported the Coix Seed has the funtion of anti-tumor and inhibit the growth of blood vessels .The purpose of the study is to observe the effect of Coix Seed extracting solution eyedrops and sub-conjunctival injection on rat cornea neovascularization(CNV),and to explore the expressions of vascular endothelial growth factor(VEGF) in CNV, to provide an effective treatment for cornea alkali burns. Methods: 80 Wistar rats were selected for experiment,5 were common contrast,Corneal alkali-burned model was established in the other 75 left-eyes of Wistar rats:Group A(control group) was treated by the normal salt solution .Group B( eyedrops group)was eyedroped by the Coix Seed extracting solution 4 times a day,for there weeks. Group C ( sub-conjunctival injection group) was treated by subconjunctival on the 1th,3th,5th ,7th,10thand 14th day ,for six times. The occurrence and development of CNV were observed slit-lamp microscope after corneal alkali burn, the area of CNV in corneas were calculated, immnuohistochemistry was used to investigate the distribution of vascular endothelial growth factor (VEGF) in rat cornea at different time.Results: The areas of CNV in group B,C was smaller than group A (P<0.05),and there was sinificant difference between group Band C(P<0.05). Immunohistochemistry showed that the positive cell number of VEGF in group B,C was lower significantly than group A (P<0.05),group B was stronger significantly than group C.Conclusion: Coix Seed extracting solution can effectively inhibition the rat corneal neovascularization(CNV) after alkali burned.
