Objective In this experiment, the five groups conduit valved was sterilizated under the different water bath conditions, through the observation of the Sterilization effect and the glucose metabolic rate and the non-Typlan's dye rate of pre-sterilization and post-sterilization,discusses the relationship between conditions water bath and tissues.On the premise of the sterilization effect, ground keep its viability as possible as can.in order to perfect the preparation method of the conduit valvedMethods This experiment selected 50 sheared conduit valved of New Zealand big white rabbit,including 25 valved arteries and 25 valved pulmonary arteries, and were randomly divided into 5groups,each group had 10 conduit valved,and was germicultured in both pre-sterilization and post-sterilization.Each group conduit valved was sterilized in the M199 solution containing Polymycin B 100mg/L,Vancomycin 50mg/L,Cefoxtin 240mg/L,Lincomycin 120mg/L,the group A were bathed in 37℃for 12h, the conduit valved in group B were bathed in 37℃for 6h, the conduit valved in group C were bathed in 37℃for 4h, the conduit valved in group D were bathed in 4℃for 12h, the conduit valved in group E were bathed in 4℃for 6h. The samples of after sterilization were obtained to examine the rate of glucose utilization and the non-Typlan's dye. The control group was unsterilized, and which samples were obtained to examine the glucose utilization and the non-Typlan's dye.Results (1) Culture result of 50 conduit valved before sterilization:9 were germiculture positive,2 were fungal culture positive, total positive rate was 22.2%. Cultured after sterilization:none of the conduits was positive.(2)The glucose utilization of all experimental groups were all lower evidently than the control group (Group A:2.03±0.29 mmol/L-24h, Group B:2.16±0.38 mmol/L-24h Group C:2.22±0.44 mmol/L-24h, Group D:2.20±0.58 mmol/L-24h Group E:2.34±0.65 mmol/L·24hThe control group:2.64±0.66 mmol/L·24h,p<0.05.and more than0.89 mmol/L·24h.(3)In the all experimental groups, The glucose utilization of group E are higher than groupA,B,C,D,P< 0.05. The glucose metabolic rate of group B and D there was no significant differences, P>0.05; The glucose metabolic rate of group C and D there was no significant differences, P>0.05.(4)The glucose metabolism rate in A, B and C three groups ware gradually heighten, P<0.05. (5)The non-Typlan's dye rate of the experimental groups compared with the control group(Group A:83.1%, Group B:87.6%, Group C:88.5%, Group D:86.3%The control group:94.0%) P<0.01. The non-Typlan's dye rate of groupE (90.6%) compared with the control group (94.0%) P>0.05,there was no significant differences,Conclusion (1)The antibiotic formula used in this experiment can obtain the satisfying sterilizing effect.(2)In this experiment, sterile conditions: 37℃water bath for 12 hours,37℃water bath for 6h,37℃water bath for 4 h, 4℃water bath for 12h,4℃water bath for 6h,can obtain sterilization effects. And keep the good cell activity.(3)The sterilizing process using antibiotics can damage the viability of the conduit valved. terilization time and temperature of water-bath to cell activity is influenti. Sterilization should minimize the time and bath temperature. In this experiment, Under the 4℃water bath for 6 hours get the less damage, and can achieve satisfactory sterilization.
