ObjectiveTo study the the changes and duration of autophagy after Subarachnoid Hemorrhage in rats, and explore the roles of autophagy in the process of SAH, whose findings may provide us a new therapy target or method.Methods20 Sprague-Dawley rats were divided into a normal group and a SAH group at randomly for the test of autophagosome. Another 40 Sprague-Dawley rats were put into normal group and SAH groups at 6h,24h,72h for the investigation of the expression of LC3 and Beclin-1. vascular insider puncture method was used to set up SAH model. The formation of autophagosome was observed by transmission electron microscopy. Western blot was performed to investigate the expression of the LC3 and Beclin-1.ResultsSAH group:autophagosome could be observed, the quantity of lysosome was increased, karyotype was irregular mostly and the endoplasmic reticulum was swollen. Control group:all organelles were normal. western blot shows that autophagic concerning protein LC3 was increased in 24h after SAH, and kept positive in 72h and Beclin-1 was increased in 6h after SAH, and kept positive in 24h.ConclusionsAutophagy is activited after SAH in rats, and could last 24hours at least. Autophagy may play a key role in the process of the SAH in early stage.
