| ObjectiveTo study the expression of the Nectin-4 mRNA and protein in ovarian cancer tissues,ovarian benign tumor tissues and normal ovarian tissues, and to investigate the relationship between the expression of Nectin-4 in epithelial ovarian cancer with the clinicopathological parameters.MethodsThe expression levels of Nectin-4 gene was evaluated by using RT-PCR and immunohistochemistry in normal ovarian tissues,ovarian benign tumor tissues and ovarian cancer tissues. And we analyzed the relationship between the Nectin-4 and the clinicopathological parameters of ovarian cancer.ResultsThe results showed the exact biological effects of Nectin-4 in ovarian cancer tissues.The tendency of two methods was coincidence on the whole.There was positive expression of Nectin-4,and the expression of Nectin-4 mRNA was observed in 0.71±0.12,0.32±0.09 and 0.30±0.09 of malignant ovarian tumors,benign ovarian tumors and normal ovarian tissue respectively, the expression of Nectin-4 protein was observed in3.35±0.52,1.36±0.41,1.15±0.44 of malignant ovarian tumors,benign ovarian tumors and normal ovarian tissue respectively.The expression of Nectin-4 in ovarian cancer was significantly higher than it in normal ovarian tissues and benign tumors (P<0.01).Statistical analysis revealed that the expression of Nectin-4 gene was not related to ages and histological subtypes (P>0.05). There was significant difference between G1, G2 and G3 (P<0.05). Close positive relation was observed between the expression of Nectin-4 protein and tumor surgical-pathological staging (P< 0.01). There was also significant difference between expression levels in tissues with lymph metastases and tissues without lymph metastases (P<0.01). ConlusionNectin-4 was highly expressed and positively correlated with ovarian cancer,but it was expressed in normal ovarian tissues negatively. The changes of the expression of Nectin-4 gene is correlated with tumor differentiation degree, Surgical-pathological staging and the metastasis of lymph node. |
