ObjectiveTo investigate the relationship of methylation in promoter of human runt-related transcription factor 3 (Runx3) gene on metastasis and oncogenesis of gastric carcinoma.MethodsMethylation specific PCR(MSP) was used to detect methylation of Runx3 promoter region of 40 gastric cancer patients (tumor tissues,adjacent tissues) and 40 gastritis patients.ResultsIt was found that the ratio of methylation in gastric carcinoma specimens is 45 percent,that in adjacent tissues is 7.5 percent, the Runx3 promoter methylation were detected in gastric carcinoma groups markedly. A significant difference was noted between two groups (P<0.05). The Runx3 promoter methylation in gastric carcinoma is correlated to character of clinic pathology,the ratio of methylation is increased along with clinic staging,but it is independent with age, gender,location of carcinoma. No methylation of Runx3 promoter was found in 29 inatrophia gastritis samples of the 40 gastritis patients But it was found in3 cases in 11 atrophia gastritis specimens of the 40 gastritis patients. A significant difference was noted between two groups(P<0.05).ConclusionRunx3 promoter methylation was one of reasons which induced Runx3 gene inactivation,it has the specificity of gastric carcinoma possibly and is correlated to on cogenesis, metastasis of gastric carcinoma.
