Objective To investigate the expression of hWAPL gene in Cervical Intraepit-helial Neoplasia (CIN) and Cervical squamous carcinoma and its significance.Mathods Collect 74 samples which obtained from patients who were cured in liaocheng people's hospital by the year 2004-2006.Take 40 Cervical squamous car-cinoma samples for the study:According to tumor type,ncluding cauliflower 19cases, tubercle 15 cases and ulcer 6 cases;According to FIGO, I stage 14 cases, II stage 15 cases,Ⅲ—Ⅳstage 11 cases;According to histological grade,Gl 17 cases,G2 10 cases,G3 13 cases;Take 19 CIN samples,including CINⅠ8 cases, CINⅡ-Ⅲ11 cases;Take 15 normal cervix tissue samples as control group.All samples haven't accept chemotherapy or radiotherapy.The hWAPL mRNA expression in different sample were detected by semi-quantitive reverse transcription-polymerase chain reac-tion(RT-PCR).Using streptavidin-biotin peroxidase(SP) method of the immunohistoch-emistry,we also examined the expression of the proteins hWAPL,HPV(Human papill-oma virus)E6,HPV E7 in above samples.Statistical analysis was performed by using SPSS10.0 statistics sofeware.Results 1.Expression of hWAPL mRNA:In normal cervix,CIN and cervical sq-uamous carcinoma,there are significane difference of mase-expression rate and expr-ession level.It is higher in CIN and cervical squamous carcinoma than normal tiss-ue(P<0.05);There are high levels in cervical squamous carcinoma than CIN and normal tissue(P<0.05).In cervical carcinoma,the hWAPL mRNA express advanced accord to the FIGO,histological grade.The expression level of hWAPL mRNA has no association with age and tumor type.2.The protein of hWAPL:The expressions in normal tissue CIN and cervical squamous carcinoma are 20%,42.2%,70%,There are significant difference(P<0.05). In carcinoma tissues,FIGOⅠstage 42.9%,Ⅱstage 73.3%,Ⅲ-Ⅳstage 100%there signi-ficant difference betweem two of them(P<0.05).According to histological grade G1 52.9%,G2 60.0%, G3 100%.There are significant difference between two of them(P<0.05).That informs hWAPL protein have positive correlation with FIGO stage and histological grade.It has no association with age and tumor growth type.3.Correlation of hWAPL protein and HPV16 E6 protein:The expression of HPV 16 E6 protein in above tissues are 6.7%,31.6%,51.2%there significant difference (P<0.05);In carcinoma tissue with HPV 16 E6 express positive and negative,the hWAPL protein positive expression are95.2%,57.9%,there are significant difference (P<0.05),indicated that expression of hWAPL protein have correlation with HPV 16 E6 protein.4. Correlation of hWAPL protein and HPV16 E7 protein:The expression of HPV16 E7 protein in above tissues are 0,36,8%,57.5%; there significant differ-ence(P<0.05);In carcinoma tissue with HPV16 E6 express positive and negative, the hWAPL protein positive expression are91.3%,58.8%,there are significant differe-nce(P<0.05),indicated that expression of hWAPL protein have correlation with HPV 16 E7 protein.Conclution 1. we detect low expression of hWAPL in normal cervix,and high expression in CIN and cervical squamous carcinoma.And the hWAPL expre-ssion may play a significant role in cervical carcinogenesis and tumor progressin.2. The hWAPL expression show a significant correlation with severity of dyspl-asia in cervical squamous carcinoma.3 hWAPL expression have correlation with HPV oncoprotein E6 and E7, informs that hWAPL may participate in the process of cervical carcinogenesis indu-ced by high risk HPV.4. hWAPL protein may be one of detection for diagnosis of cervical squamous carcinoma.
