Objective: To investigate the protective effects of Panax quinquefolium saponins from steams and leaves (PQS) on focal cerebral ischemia damage and its mechamism.Method:The male rats were pretreated with PQS 50 and 100㎎·㎏ -1 ig, one time per day for 7d, and then subjected to focal cerebral ischemia damage induced by a middle cerebral artery occlusion(MCAO) for 24h. Determined neurological symptom by Longa's score at the point 0h,24h after cerebral ischemia, and then drawn, brain tissue pathological changes was observed by light microscope; and using enzyme-linked immunosorbent assay (ELISA) test to detect the contents of NGF and TGF-β1 seperately in ipsilateral brain tissue.Results:1.PQS can significantly improve the focal cerebral ischemia in rats signs of neurological deficit, reduced neurological deficit scores, reduce the pathological changes of ischemic brain tissue.2.PQS can significantly increased the content of NGF in rat brain after focal cerebral ischemia.3.PQS can significantly increased the content of TGF-β1 in rat brain after focal cerebral ischemia.Conclusions: PQS has protective effects on focal cerebral ischemia damage and its mechanism may be related to inducing the expression of NGF and TGF-β1.
