Objective By establishing the models of liver fibrosis in rats induced by dimethylnitrosamine(DMN), to investigate pathologic changes of the hepatic tissue and hepatic fibrosis scoring changes in SSS, Using Reverse transcription-polymerase chain reaction(RT-PCR) to detect renin-angiotensinâ…¡(Angâ…¡) 1 receptor (AT1R) mRNA and angiotensin-converting enzyme (ACE) mRNA expression in rat liver fibrosis in the process of dynamic change, and inquiring into the prophylactic effect of compound taoheshenshu on experimental liver fibrosis in rats induced by DMN.Methods With DMN-induced rat liver fibrosis model, rats were given 1% of the DMN (1 ml/kg,1 time/day for 3 consecutive days per week for 4 weeks) were injected intraperitoneally. Rats of the control group were given the compound taoheshenshu at the same time. Liver tissue samples were examined on 14,28,42days.Each group was observed in rat liver histopathological changes, calculation of hepatic fibrosis of rat liver tissue SSS scoring and used RT-PCR assay AT1RmRNA and ACEmRNA expression.Results 4 weeks after modeling, typical liver fibrosis formed,and 6 weeks after modeling, the hepatic injury in the model group had some remission as compared with that at 4 weeks. Compared with model group, Control group of rat liver tissue fibrosis were significantly improved, SSS score of liver fibrosis were significantly lower (P<0.05), AT1RmRNA expression level was significantly lower (P<0.05 or P<0.01), ACEmRNA expression level was significantly lower (P<0.05).Conclusion 1.Intraperitoneal injection of DMN solution lml/kg,1 time/day for 3 consecutive days per week for 4 weeks can be established the model of liver fibrosis successfully.2.The compound taoheshenshu can improve the DMN-induced liver fibrosis in rats and reduce SSS scoring.3.The compound taoheshenshu can reduce the expression of AT1RmRNA and ACEmRNA.
