Background and objective As a kind of basic immune depressants, TWP is applied in clinic generally including renal glomerular disease, rheumatoid arthritis , anti-exclusion of organ transplant and so on. It has a bright future in clinical application, but it's adverse effects confine clinicial application especially its disadvantage of genital system. According to statics, more than half women who use TW over half a year can appearance menstrual disorder or amenorrhea. Aimed at impairment of genital system induced TW, we used Estrogen- Progestogen sequential replacement therapy in clinic and gained good consequence . To test our hypothesis ,we design a suit of empirical method and expect to use this therapeutic regimen widespreadly in clinic and the study of drug.At the same time,some researchs indicate that to test the espression of foxl2 mRNA on ovary granulosa cell(GCs) can directly reflect the stored function of ovary, also can reflect the functional status of ovary as a earlier monitoring target.Objective To investigate the protective effect of estrogen combined with progestogen replacement therapy on the femal rats with the impaired of genital system induced by Triptergium wilfordii polyglycosidium and to approach the possible mechanism on the basis of the expression of foxl2 mRNA on GCs.The 60 SD femal rats were equally randomized into three groups: control groups,recover groups and treatment groups. The process lasted 16 weeks. The effect of TWP and HPT was evaluated by change of estrous cycle, humid weight of ovaries and uterus in female rat, change of histological specimen with ovaries and uterus and the levels of E2, LH and FSH in their blood. The expression of Foxl2 mRNA were detected by RT-PCR on ovary granulosa cell.Result Compare with the control groups, TWP can decrease the quantities of rats with nomal estrous cycle, and the humid weight of ovaries and uterus. The level of E2 secreted by ovary was lower and the level of LH,FSH secreted by pituitary was higher after treating by TWP than those of control groups .there was significant difference in control groups and TWP groups (P<0.05) . Estrogen combined with progestogen replacement therapy can restrain these pathological changes produced by TWP, and there was not significant difference in control groups and healing groups(p>0.05). Conclusion TWP can damage the genital system of femal rats. It is an effective method that treating by estrogen combined with progestogen replacement therapy periodically in these rats.
