Objective:To synthesise a new bone repair material which was composed of chitosan and phosphorylated chitosan by biomimetic method and to evaluate its cytocompatibility in vitro.Methods:Phosphate group was incorporated into chitosan to form phosphorylated chitosan.The phosphorylated chitosan solution was dropped into the acetic acid solution of chitosan, and then dry the sediment to get the complex of chitosan and phosphorylated chitosan. CS/PCS hydrocolloid was co-cultured with rat osteoblast in vitro to observe the adhesion and growth of osteoblast.Hydroxyapatite group was using as control group.The proliferation and differentiation of osteoblast was determined by MTT and alkaline phosphatase methods.Results:1.The morphological change of CS/PCS was from unstable to stable.2.The transverse plane observation of CS/PCS showed it was porous reticular and the diameters of holes were about 100-200nm.3.The osteoblasts response to the mineralized CS/PCS material showed that it was considerably cytocompatible, which can promote osteoblast proliferation and differentiation. Conclusion:The biomimetic CS/PCS composite had porous structure. It has well cytocompatibility after mineralizing in saturated Ca(OH)2 solution. CS/PCS composite can promote proliferation and differentiation of osteolast.It can be used as a new bone repair material.
