Objective: To investigate the role of CD40 in Colon cancer by examining the expression of CD40 and its significance in colon cancer.Methods: (1) The expression of CD40 was examined by immunohistochemistry in human colon cancer tissues(57cases) and adjacent tissues(57cases). (2) 44 fresh colon cancer cells were examined by RT-PCR. (3) 22 fresh colon cancer cells were examined by FCM (flow cytometry) . (4) The correlations of CD40 molecule expresstion with clinic stage ,histologic types, pathologic grades and metastasis of colon cancer were alsoanalyzed by usingχ2 test .Results: (1) The expression rate of CD40 in colon cancer tissue was 42.1% (24/57) ,.and 9.2% in adjacent tissue (P<0.01) . (2) CD40mRNA was examined by RT-PCR in colon cancer tissues and adjacent normal tissues from 44 cases. The positive rate of CD40 was 43.2%(19/44) in colon cancer tissue and 11.2% in adjacent tissue.(3) The expressive contents of tumor cells and adjacent normal tissues were comparatively studied by flow immunocytometry in 22 cases with colon carcinoma. The CD40 expressive contents of colon carcinomas were 40.9%(9/22),more than 4.5%(1/22) of normal tissue(P <0.01). (4)A statistically significant correlation was found between the expression of CD40 (CD40mRNA) and lymph node metastasis. CD40 and CD40mRNA expression showed no significant relationship with tumor size.Conclusion: CD40 molecule might play an important role in the carcinogenesis of colon cancer and might be a valuable marker in predicting the prognosis of the colon cancer. The high expression of CD40, alone and in combination, promote the invasion and metastasis of colon cancer.It is significant for the therapy of the patients to anticipatethe prognosis of colon cancer.
