Objective:To investigate the anti-angiogenesis effects of the combin-ation of low-dose Docetaxelc (TXT) and Ginsenoside Rg3 in nude mice with human breast cancer, and to observe the anti-tumor effect.Method:1.MCF-7 human breast cancer cells(6×106) suspended in 100μl of DMEM-50% Matrigel were injected into each flank of nude mice 2.Four groups were randomly divided:Ginsenoside Rg3 group,low-dose docetaxel group,combination group,control group,receievd Ginsenoside Rg3 (5mg/kg,every other day,intraperitoneal injection,continuous 21d), TXT(0.5mg/kg every other day,intraperitoneal injection,continuous 21d),and no rmal saline(same volume,every other day, intraperitoneal injection, continus 21d).respectively.3.Tumor volume and write blood cell count of nude mice were measured in each group every week.4.All nude mice were killed until to the 21th day,then took the tumors.All tumors were embed by 10% formaldehyde solution respectively. Immunohistochemistry SP(Strepatacidin peroxidase System For Immunohistological Staining)was used to detect the expressions of vascular endothelial growth factor (VEGF) and microvessel density (MVD) and matrix metalloproteinase-9(MMP-9) in different tumor tissues.Result:1.All the mouses were at good condition before experiment and all were alive after it.2.Through dynamic observation,control group mouses were obvious maransis with less activites.Interfered group mouses forage,drink and act normally without diarrhea and congestion.3.The difference in the volume of all the tumor grafts were not statistically significant when theapy started (P>0.05);the tumor grafts appeard to grow slowly of nude micein Ginsenoside Rg3 group and low-dose docetaxel group and combination group after therapy 7days,compared with control group respectively were statistically significant(P<0.05);the growth rate of tumor grafts in combinaion group was significantly slower than that of the other 3 groups (P<0.05) the difference of the volume of tumor grafts in 4 groups were significantly at the end of experiment(21d) (P<0.05).4.In Ginsenoside Rg3 group low-dose docetaxel group,combination group,control group the data showed that Weights of tumor with antitumor rate were 1.175±0.05g, 38.96%;15±0.041g,40.26%;0.95±0.058g,50.65%;1.925±0.171grespectively; The treatment groups tumors were lower in weight than those in control group(p<0.05) and the combination group tumors were lower than those in monotherapy groups (p<0.05)5.The difference in the write blood cell of four groups were not statistically significant (P>0.05).6.The expression of VEGF and MMP-9 and MVD of treatment groups tumors were lower than those in control group(p<0.05)and the expression of combination group were lower than those in monotherapy groups (p<0.05).Conclusion:Ginsenoside Rg3 and low-dose docetaxel can inhibite the growth of breast cancer through anti-angiogenesis.The combinating therapy of Ginsenoside Rg3 with low-dose docetaxel shows more marked inhibition on the growth of breast caneer. without obvious toxic side effect.It may be applied in combination chemotherapy of breast.
