Objective:Observing the growth of tumor,toxicity and adverse reaction in the mice with sarcoma s-180 and the change of biological marker which reflect angiogenesis,proliferation of tumor treated by combination of ginsenoside Rg3 and low-dose cyclophosphamide(CTX).To investigate the mechanism of them and establish associated testing with tissue and in order to put it in clinical work and kinesis observation.Methods:BALB/c sarcoma s-180 mice were taken as the model and randomly divided into 4 groups,i.e.the model group and the experimental groups that was treated with ginsenoside Rg3,or low-dose CTX,or low-dose CTX combined with ginsenoside Rg3 in mice,then detect markers like followings:1.To observe tumor volume and weight of mice.2.To examine tumor tissue,such as vascular endothelial growth factor(VEGF), microvascular density(MVD)and proliferating cell nuclear antigen(PCNA)by immunohistochemistry.Recult:1.In the LDCTX group,Rg3 group,LDCTX+ Rg3 group,tumor grew comparatively slow than model group(P<0.05),the combinational group tumor volume and weight also was significant less than those in individual treatment therapy groups(P<0.05).2.In the LDCTX group,Rg3 group,LDCTX+ Rg3 group,the expression of VEGF and MVD decreased compared to model group(P<0.05),and the lowering action was more significant when the two drugs were used in combination (P<0.05).In the Rg3 and the combinational group tumor tissue expressed more PCNA than model group(P<0.05).Positive correlation was found between the expression of VEGF,MVD and PCNA of tumor tissue.3.The combined therapy of CTX and ginsenoside Rg3 resulted in better quality of mice life.Conclusions:1.The ginsenoside Rg3 or low-dose CTX can inhibit tumors'growth and diminish expression of VEGF protein and MVD.The combination of low-dose CTX and ginsenoside Rg3 has obvious synergetic action of anti-angiogenesis.2.The ginsenoside Rg3 showed inhibitory effect on the cell proliferation of sarcoma s-180. 3.The ginsenoside Rg3 can improve survival quality and reduce toxicity in imce with sarcoma s-180.
