SDF-1-CXCR4 Biological Axis In Renal Cell Carcinoma Metastasis

Objective:To investigate the relationship between SDF-1-CXCR4 biological axis and renal cell carcinoma metastasis, by detecting difference of CXCR4 expression between metastatic renal cell carcinoma and non-metastatic renal cell carcinoma expression and the difference of SDF-1 expression between lymph node and ureteral.Methods:To select metastatic renal clear cell carcinoma specimens of 32 cases, non-metastatic renal clear cell carcinoma specimens of 31 cases. Among them males 42 cases, females 21 cases; less than 50-year-old 16 cases, older than or equal to 50-year-old 47 cases; pathological Fuhrman grade,Ⅰgrade 7 cases,Ⅱgrade 39 cases,Ⅲgrade 13 cases,Ⅳgrade4 cases. To detect the difference of CXCR4 expression between metastatic renal cell carcinoma and non-metastatic renal cell carcinoma expression by immuno-histochemistry. And to detect the sex, age, Fuhrman grade difference in CXCR4 expression. To analysis the results by semi-quantitative immuno-histochemistry.To select lymph node specimens of 30 patients and ureter specimens of 30 patients.All the people are renal cell carcinoma patients. Among them males 46 cases, females 14 cases; less than 50-year-old 19 cases, older than or equal to 50-year-old 41 cases; pathological Fuhrman grade,Ⅰgrade3 cases,Ⅱgrade 36 cases,Ⅲgrade 17 cases,Ⅳgrade 4 cases. To detect the difference of SDF-1 expression between lymph node and ureteral. And to detect the sex, age, Fuhrman grade difference in SDF-1 expression. To analysis the results by semi-quantitative immunohistochemistry.Results:In 32 patients with metastatic renal cell carcinoma CXCR4 expression score was 126.72±74.72. In 31 cases of non-metastatic renal cell carcinoma CXCR4 expression score was 63.39±60.69. The t test, P<0.05, significant statistically difference between the two things. In 42 cases of male patients CXCR4 expression score was 91.55±77.15, In 21 female patients CXCR4 expression score was 103.57±71.29.The t test, P> 0.05, no statistically difference between the two. In 16 cases of less than 50 years of age CXCR4 expression score was80.00±76.25.In 47 cases of greater than or equal to 50 years old CXCR4 expression score was 100.85±74.50. The t test, P> 0.05, no statistically difference between the two.In 46 cases of Fuhrman gradeⅠ+Ⅱlevel CXCR4 expression score was97.83±76.73,In 17 cases of Fuhrman gradeⅢ+Ⅳlevel CXCR4 expression score was 89.41±71.54. The t test, P>0.05, no statistically difference between the two.In 30 cases of lymph node SDF-1 expression score was 145.00±46.42,In 30 cases of ureteral SDF-1 expression score was 63.50±23.49. P <0.05, significant statistically difference between the two. In 46 cases of male patients SDF-1 expression score was 99.02±55.56, In 14 female patients SDF-1 expression score was 121.43±51.00.The t test, P> 0.05, no statistically difference between the two. In 19 cases of less than 50 years of age SDF-1 expression score was 104.21±49.08. In 41 cases of greater than or equal to 50 years old SDF-1 expression score was 104.27±58.05. The t test, P>0.05, no statistically difference between the two.In 39 cases of Fuhrman gradeⅠ+Ⅱlevel SDF-1 expression score was 100.64±56.10,In 21 cases of Fuhrman gradeⅢ+Ⅳlevel SDF-1 expression score was 110.95±53.42. The t test, P>0.05, no statistically difference between the two.Conclusions:CXCR4 expression in metastatic renal cell carcinoma was higher than in non-metastatic renal cell carcinoma. In other words, the higher intensity of CXCR4 expression in renal cell carcinoma is more prone to metastasis. CXCR4 chemokine receptor expression may be associated with renal clear cell carcinoma metastasis.SDF-1 expression in Lymph nodes was higher than in ureter. The organization with higher SDF-1 expression is more prone to metastasis. SDF-1 may be associated with renal cell carcinoma metastasis.SDF-1-CXCR4 biological axis is closely related with the renal cell carcinoma metastasis.
