The test researched the inhibition of Chinese traditional compound medicine"LN" (Refined and crude) in subinhibitory concentration to the multidrug resistance of the E.coli to Tetracycline, chloramphenicol and quinolones category by the test of culture in vitro. The drug resistance gene AcrA of the E.coli was amplificated by the polymerase chain reaction (PCR). Detecting the influence of Chinese traditional compound medicine"LN" (Refined and crude) to the drug resistance gene AcrA of the E.coli by inactive test of drug resistance gene and PCR, then discussed the mechanism of action of reducing AcrA on drug tolerance to the multidrug resistance of the E.coli by"LN" (Refined and crude)Adopt Real-time PCR quantitive detection drug-resistance gene AcrA-mRNA of each E.coli before and after by dealing with the Chinese traditional compound medicine"LN" (Refined and crude), and contrasting and analyzing drug-resistance gene AcrA-mRNA of each drug-resistance E.coli, study on the influence of drug-resistance gene AcrA-mRNA of E.coli by, probe the mechanism of action of reducing AcrA on drug tolerance to the multidrug resistance of the E.coli by"LN" (Refined and crude)Result:Traditional Chinese medicine with yellow (refined to mention crude extract) on the inhibition rate of DLl-resistant Escherichia coli K88 compared with the susceptible ones was no significant difference (P>0.05),the inhibition rate of the NUT and the K88 significant difference (p<0.05).And refined to mention preparation and crude extract preparation was significant difference (p<0.01).Contrasting the common relation of base sequence of AcrA and amino acid sequence between the drug resistance E.coli DL1 which was amplificated by the polymerase chain reaction (PCR) and ECU00734 which had been reported in Genbank, the result was 100%; it inditated that we have amplificated drug-resistance gene AcrA of E.coli; contrasting the common relation of base sequence of AcrA and amino acid sequence between the drug resistance E.coli DL1 which was amplificated by the polymerase chain reaction (PCR) and ECU00734, the result was 99%; contrasting the common relation of base sequence of AcrA and amino acid sequence between the drug resistance E.coli NUT and ECU00734, the result was 98%.The bases in 608-693 changed largely before and after dealing with by the Chinese traditional compound medicine "LN":(Refined and crude), and the propotional amino acid sequence was also changed, it appearanced amino acids loss. Detecting quantitive the mRNA expression of the drug resistant gene AcrA in different of the strain DL1 (after), NUT(after),by Real-time PCR and compareing with propotional strain before dealing with by LN. And refined than the crude preparation to mention agents of the AcrA mRNA expression levels of greater impact, mRNA expression levels of lower their more obvious.Conclusion:The Chinese traditional compound medicine"LN" (Refined and crude) in subinhibitory concentration can reduce the drug tolerance of E.coli to Tetracycline, chloramphenicol and quinolones category antibiotics effectually; the mechanism of action that is due to change a part of bases of the drug resistance gene AcrA, then the drug resistance gene is inactivated. So that AcrA gene mRNA expression levels change-The Chinese traditional compound medicine"LN" (Refined and crude) in subinhibitory concentration could effect the course of expression of the drug resistance gene AcrA.
