Part onePurpose:The purpose of this study was to observe the expression of DKK3 and vWF protein in colorectal carcinoma and glioma and their clinical significance, to nvestigate the correlation between DKK3 and vWF and the association between the two factors and MVD.Methods:IMH staining was used to detect the expression of DKK3,vWF protein in colorectal carcinoma tissue microarrays containing 94 colorectal carcinoma specimen and 31 glioma.Results:The expression of DKK3 in colorectal carcinoma was lower or disappear than that in para-cancer mucosa, expression of DKK3 was not found in vascular endothelial cells; The expression of v WF in colorectal carcinoma was higher than that in para-cancer mucosa;The expression of vWF was found in colorectal carcinoma and glioma; The expression of DKK3 and vWF in colorectal carcinoma was not correlated to age and sex of patients, invasive depth and tumor locus of colorectal carcinoma,except differentiation, pTNM stage and lymph-node metastasis;The expression of DKK3 and vWF in glioma was not correlated to age and sex of patients, histological grouping of glioma, except clinical stage;The expression of DKK3 and vWF in colorectal carcinoma and glioma was not correlated to MVD;The expression of DKK3 was not correlated to the expression of vWF in colorectal carcinoma and gliomaConcluslon:DKK3 may not be involved in the tumor angiogenesis; DKK3 could be involved in inhibiting tumor metastasis;May be vWF involved in the tumor metastasis; The expression of DKK3 was not correlated to the expression of vWF in colorectal carcinoma and glioma; suggesting that vWF and DKK3 playing different roles in different stages of development in tumor. Part twoPurpose:To nvestigate the expressions in human colon carcinoma cell line SW480 and effects on the growth adhesion, invasion and motility of DKK3 and COX-2 in human colon carcinoma cell line SW480 in vitro. After tumor cells of SW480 and HUVEC were co-cultured, expression of DKK3 and capillary formation of human umbilical vein endothelial cells was observed.Methods:Human colon carcinoma cell line SW480 cultured in vitro were treated with DKK3 antibody(DKK3Ab).The expressions of DKK3 in SW480 and HUVEC were detected by ICM and IF.The effects of DKK3 respectively and combine COX-2 on the proliferation, adhesion, invasion and motility of human colon carcinoma cell line SW480 in vitro were explored respectively by MTT colorimetric assay,Boyden chamber and wound healing assay.After tumor cells of SW480 and human umbilical vein endothelial cells were co-cultured, the expression of DKK3 and capillary formation of human umbilical vein endothelial cells was investigated by IF and HE.Results:DKK3 was expressed in the human colon carcinoma cell line SW480; Compared with the control group,the growth,adhesion,invasion and motility abilities of the SW480 cells were increased significantly after treatment with DKK3 antibody; Compared with DKK3Ab treatment group, the adhesion, invasion and motility abilities of the SW480 cells decreased significantly after treatment with DKK3Ab+ COX-2Ab group; DKK3 was not expressed in HUVEC; After tumor cells of SW480 and HUVEC were cultured, the expression of DKK3 and vascular ring formed of HUVEC which was changed by DKK3Ab was no changed.Conclusions:DKK3 protein was expressed in colon cancer cell line SW480 cells; DKK3 may one of tumor suppressor gene; COX-2 may be have correlation with DKK3. The expression of DKK3 was not expressed in human umbilical vein endothelial cells, DKK3 had no effected on the formation of vascular rings human umbilical vein endothelial cells. |
