carbohydrate, which is extensive in nature and organism, is the important nutrients for the human body. However, the new study found that carbohydrate not only can provide the energy but also has some other functions, such as anticancer, antibacterial, antiviral, regulating cell growth and human immune. Therefore, people give more and more attention to carbohydrate.Because biological molecules such as sugar has only weak UV absorption, it is mainly determined through the following methods at present: chromogenic reagent-sulfuric acid method, DNS method, titration method, paper chromatography, TLC, GC, HPLC and so on. However, these methods comsumed a long analysis time besides poor precision and quantification. For this reason, we involved the use of HPLC-MS method to detect the sugar components, which is of great significance in detection of the carbohydrate.In recent years, with in-depth understanding of people's capabilities for sugar, sugar aroused people's interest in the study of it, such as springing up a hit. Among them, sugar structure is of critical importance. As Carbohydrate has different types and number, the structure of sugar is very complicated. Mass spectrometry, in particular, GC-MS (EI or CI mode) for single-sugar analysis already has several decades of history. A variety of soft ionization techniques can be effectively carried out the direct detection of carbohydrates without any derivative steps, which brings the rapid progress in carbohydrates Research.This thesis focused on those researches:1. This paper has reviewed recent research progress, currently used detection methods, and also discussed the advantage of high-performance liquid chromatography-mass spectrometry detection in the field of carbohydrate detection.2. The procedure to analyze stachyose with HPLC-ESI-MS was set up.This method optimized the conditions of separation and mass spectrometry response, which could accurately detect stachyose in Liuwei Dihuangwan and can provide strong basis for quality control because of low detection limit and high precision.3. A HPLC-ESI-MS method for deteminatioon of glucose, sucrose, raffinose and stachyose in the process of rice growth was proposed. Dynamically analyzed content changes in different types and different developmental stages of rice, we found that sugar content changed with types and growth period. This method optimized the conditions of separation and mass spectrometry response, which can be successfully used in monosaccharide and oligosaccharide detection of rice.4. The procedure to analyze carbohydrate with HILIC-ESI-MS was set up. 5.By analyzing structural of saccharide(monosaccharides, disaccharides, trisaccharide, tetrasaccharide) by ion trap mass spectrometry, we summed up the fragmentation pattern of carbohydrate.
