Heparin is a polydispersed and highly sulfated glycosaminoglycan (GAGs), it is comprised of alternating 1 -*4 lined uronic acid and glucosamine residue, its major repeating disaccharide unit is -[-a-L-IdoA-2-SO4-P(l-*4)- a-D-GlcNSO3-6-SO4-]-. It is found that heparin has anti-asthma effects, which is mainly related to its anti-inflammation effect in recently studies. The anti-asthma and anti-inflammation effects are closely related to their characteristics of polyanion and variable structure, they can bind with proteins, cytokines and oligosaccharide ligands in vivo which have positive electricity, and then, they inhibit inflammation and airway hyperreactivity. As an anticoagulant, heparin has an adverse reaction of hemorrhage, when it is degradated and sulfated, the anticoagulant activity will be lowed, and the adverse reaction will be decreased.To study the relationship between the anti-asthma effect and the structure of sulfated heparin oligosaccharides, the feasibility of three methods to degradate sulfated heparin were studied. Sulfated heparin oligosaccharide LPSH-3, LPSH-4 and heparin oligosaccharide LMWH8-1 were produced by degradation and purification, and their structures were analyzed. In order to study the anti-asthma mechanisms of sulfated oligosaccharides, real-time PCR method was used to determine the mRNA expression of IL-4, IL-5 and IL-13 in lymphocytes of asthma model mouse administered with different oligosaccharides, and immunohistochemistry method was...
