The study of this article was mainly on basis the literature research of Shengmai Yin of Chinese herbal compound and its ministerial drug dried root tuber of Ophiopogon japonicus (Thunb.) Ker-Gawl. of Radix Ophiopogonis, the purification process of the effective fraction in the compound, quality standards and chemincal composition in Radix Ophiopogonis,ect. Aimed at raising the content of sites Shengmai Yin effective; perfect the quality standards of Radix OphiopogonisIn the studiesthe of the purification process of Shengmai Yin effective parts, use Ophiopogonin D and Schizandrin for etermine the total saponins and total Lignans. And then the purity and transfer rate of the two effective parts on the Shengmai Yin purification to be monitored. Using macroporous resin for Purifyingthe purification of Shengmai Yin effective parts screening of different models of macroporous resin, the resin was investigated static and dynamic adsorption performance, and the sample concentration and flow rate, eluent concentration and volume-related factors were optimized.It was confirmed that the best purification process was AB-8 macroeticular resins, total flavonoids of crude drugs to dried resins being 1:1, eluted by 15BV 70% ethanol after 15BV water with flow rate of 1BV/h.70% ethanol part was collected and dried under vacuum. resulting in the triterpenoid total saponins content reaching 19.94%, metastasis rate reaching 36.69%; the steroidal total saponins content reaching 31.19%, metastasis rate reaching 37.59%; the total Lignan content reaching 28.62%, metastasis rate reaching 39.11%; to triterpenoid saponins, total saponin and total lignans purity 48.66%, metastasis rate wsa 38.08%; to steroidal saponins, total saponin and total lignans purity 59.81%, metastasis rate was 38.30%.In qulity standards of Radix Ophiopogonis studies, adopt three methods of identification, microscopic identification of its cross-section of the phloem, macroporous resin enrichment for TLC and identification of evaporative light scattering detector to study the differences in Radix Ophiopogonis and Radix Liriopes; use Vanillin-perchloric acid method to determine the total saponin content of 16 approved medicines With Radix Ophiopogonis saponin D as index components, saponins tentatively this calculation by dried, total saponins content shall not be less than 0.4%; With evaporative light scattering detector at the same time, the content of 16 approved Radix Ophiopogonis Ophiopogon D and Ophiopogon D'were determined, tentatively scheduled for this calculation by dried, the content of the ranges of Radix Ophiopogonis Ophiopogon D and Ophiopogon D'wre respectively 0.008%-0.029% and 0.007%-0.025%.In chemical constituents studies, pre-experiments of Radix Ophiopogonis medicinal was performed to obtain a better understanding of the specific circumstances of the chemical constituents contained.then the Radix Ophiopogonis medicinal was extacted by ethanol, respectively, enrich and purify total Ophiopogonis of its extracts by organic solvent extraction and macroporous resin for observation and comparison. It was confirmed that use two technologies which are organic solvent extraction and macroporous resin for enrich and purify total Ophiopogonis. n-Butanol extraction and the enrichment was obtained from the extaction continue to excessiving macroporous resin, and then Respectively adopt ordinary silica gel, dextran gel, reversed-phase silica gel to separate and purify, and get four compounds, including two identification with the reference substance, namely daucossterol and Radix Ophiopogonis Ophiopogon D. Compare the D. Separation Process of two enrichment parts, Results show that total saponins of pre-enrichment, there were more conducive to isolate monomers with the higher purity of total saponins and reducing the number of times on column separation.
