The Research Of HLJD Decoction On Inflammation With Atherosclerosis

Objective:This program set up the model of atherosclerosis feed with cholesterol diet combined vitiminD-3 double factors,involves the aspects of atherosclerosis through blood lipids, inflammation factors, T cell, used the treatment of HUD decoction,in order to research the effect Qirejiedu method on these indicators, found out the new cerebration for the therapy of atherosclerosis.Methods:At the first stage, divided the rats into six groups, the normal group,the model group,HLJD decoction high group, middle group, low group,set up the animal model of atherosclerosis feed with cholesterol diet combined vitiminD-3 (every 2 weeks of 200000U every time, the total to 600000U per rat) the normal group feed the normal diet for 8 weeks, at the sixth week give three groups with HLJD decoction low dosage,middle dosage and the high dosage, one group with atorvastati capills for 4 weeks, at the tenth week,the aorta and blood were prepared for the test, the blood was assayed by blood analysis machine to test the blood lipids,by ELISA method to assay intercellular adhesion molecule l(sICAM-1) and vascular cell adhesion molecule 1(sVCAM-1), by flow cytometry to measure the le-vels of CD4+CD25+Treg,the aorta was assayed by Hematoxylin and Eosin (HE)staining to investigate the effect of atherosclerotic lesion and by RT-PCR to assay the expression of C Reactive Protein(CRP), monocyte chemotactic protein-1(MCP-1)and chemokine receptor 2 (CCR2),Result:The effects on blood lipids, AS model group, CHOL, TG, LDL-C expressed higher compare to the control group, which were significantly different. The level of HDL-C sig-nificantly lower compare to the control group, there are statistically significant. Compare to the model group, the level of CHOL,TG,LDL-C were significantly lower in HLJD de-coction three treatment groups and atorvastati group, both are of significant difference.On aortic pathologicstaining, the results from the pathological examination showed that experiment copied atherosclerosis model successfully, the control group, intima thin and structural integrity. In the model control group, a great quantity of widespread fatty plaque were seen, in which intima thickened and foam cell accumulated obviously. In con-trast, giving HLJD decoction for treatment, the fat, lipid streaks and fibrous plaques has been reduced, both quantity and volume of foam cell decreased.In vascular adhesion factor, compared with normal group, the level of sICAM-1 and sVCAM-1 in the model group were significantly higher. Compared with the model control group, the expression of sICAM-1 and sVCAM-1 in HLJD decoction middle group, high group were significantly decreaed.The level of sICAM-1 and sVCAM-1 were positive correlation.The parameters of CRP was decreased, there was a significant difference before and after the treatment of HLJD decoction and atorvastati group compare to the model group, the therapeutic effect in treatment group was superior to control group.The mRNA expressions of MCP-1 and CCR2 in post transplant recipients were signifi-cantly higher compared to normal control group. Compare to the model group, the expres-sion of MCP-1/CCR2 mRNA in aortic was significantly lower in HLJD groups and ator-vastati group,the level of MCP-1 and CCR2 were positive correlation.On CD4+CD25+Treg expression of the normal control group and model group CD4+CD25+, CD4+CD25+percentages were significantly different (p<0.05), model control group CD4+,CD25+,CD4+CD25+was reduced, administration intervention, HLJDD group CD4+CD25+percentage significantly higher than the model group (P<0.05).Conclusion:HLJD decoction has the significant effect on the therapy on atheroscle-rotic, which of the efficacy on the regulation of blood lipids, pathological changes, and low down the expression of inflammatory factor, upgrade the level of T cell, also for the better of immunity function.
