IntroductionTumor metastasis is a important reason which lead to therapy failure and death, Several tumor metastasis associated genes were clonedin recent years,MTA(Metastasis associated gene)family is one of them.Metastasis-associated gene 2.MTA2 is a member of The MTA family,MTA2 expresses scarcely in normal tissue,while it's level of expression always increases in pathology state.The generality of it's expression in cancer is to be investigated.The expression of MTA2 in Non-small Cell Lung Cancer and the relationship between MTA2 and clinical pathology characteristic of NSCLC is seldom studied at present.Therefore,In this test we design to examine the expression of MTA2 in non-small cell lung cancer and analyze the relationship between the expression of MTA2 and NSCLC clinical pathology characteristic.Materials and Methods1.Tissue samples110 NSCLC with 34 their neighboring cases of noncancerous tissue samples were obtained from patients underwent surgical resection in the First Affiliated Hospital of China Medical University between 2006 and 2007.All were fixed in formaldehyde, embedded in paraffin.2. ReagentsThe primary antibody anti-human MTA2 goat polyclonal antibody were produced in Santa Cruz Co. DAB agent kit and horseradish peroxidase labeled rabbit-anti-goat IgG were bought from Beijing Zhongshan Golden Bridge Biotechnology Co.S-P kit were purchased from Fuzhou Maixin Biocompany.3. MethodsImmunohistochemical staining was performed using ant-MTA2 antibody(1:100) according to S-P method. The sections were deparaffinized, water processed, treated by high tempreture to expose antigen and incubated with primary antibody at 4℃overnight. The color was developed with diaminobenzidine and the section were couterstained with harmatoxylin.The expression of MTA2 by staining was graded for the percentage of immunoreactive tumor cell, After the areas of tumor tissue had been chosen by random under low power, one hundred cells within tumors were counted in each of 10 fields at 200 magnification. The case which the masculine percentage esceeded 10% was positive one.4 Statistical analysisThe statistical software SPSS 17.0 was applied for data analysis.pearson correlation test was used to study the relationship between MTA2 expression and clinicopathological characteristics. P<0.05 was significative in statistic.Results1. Expression of MTA2 in NSCLC and it's neighboring noncanceious tissue.There are 64 cases of NSCLC tissue that appeared yellow or even brown particles in nucleus of tumor cell among 110 cases, and in epithelia of bronchi or alveoli in neighboring noncancerous tissue. MTA2 protein showed negative expression.2. The relationship between the expression ofMTA2 and pathological features. The expression of MTA2 was correlated closely with differentiation degree and clinic stage and lymph node metastasis, but had no relationship with age. gender, historical type.DiscussionThe recent research found that MTA2 was a subunit of nucleosome remodeling and histone deacetylation compound., which may be play a key role in the course of tumor metastasis. It was gradually reported that MTA2 was overexpressed in breast cancer, ovarian cancer and liver cancer, and In addition, the level of MTA2 expression was higher in metastatic lesions in lympha nodes derived from ovarian cancer than in primary tumor and normal epithelia. Our experiment showed that MTA2 was overexpressed in non-small cell lung cancer, which is positive associated with lymph node metastasis and clinic stage. This result implies that MTA2 can promote the growth and metastasis of NSCLC cell.Conclusion1,MTA2 is overexpressed in non-small cell lung cancer, which is positively correlated with lymph node metastasis and clinic stage but negatively with ifferetiation deg ree. 2,MTA2 can promote propagation and transfer of NSCLC cell,so,it can act a arget in diagnose about NSCLC early stage.
