| ObjectiveAdenine fed rat model of chronic renal failure and the optimal dose evaluation, the establishment of norms, standards, and stable chronic renal failure rat model of adenine.MethodsBefore the experiment, randomly selected 20 rats and blood and urine initial value. Wistar rats were randomly divided into control group, A group,; model group, B group, C group, D group. A group received normal saline, the model group, groups were given different doses of adenine administered in each group were measured after 1,2 weeks of relevant indicators,3 weeks after the randomly selected five rats were sacrificed and the specimens were related to indicators, the remaining rats fed adenine stopped at 7 weeks (4 weeks after) the remaining rats were sacrificed related indicators, specimens were taken. Calculate the mortality rate of rats, blood in the abdominal aorta to detect serum creatinine, sacrificing with 24-hour urine metabolic cage, urine creatinine, urine protein, with the TUNEL method (terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling, TUNEL) test specimens of glomerular apoptotic index [1] [4].ResultsThe model rats with adenine irrigation drugs increase the number of days, to chronic renal failure, serum creatinine gradually increased. With the filling of all the model group increased the concentration of adenine, the death rate has increased.7 weeks (4 weeks after), serum creatinine has dropped substantially, withdrawal (4 weeks) before and after the existence of significant differences (P<0.05), and the C group, D group, D group and the initial group value was no significant difference (P> 0.05), while the B group, C group, D group before and after drug withdrawal was no significant difference in glomerular apoptosis index (P> 0.05).ConclusionAdenine fed rat model of chronic renal failure, the optimal dose is 200mg/mg/d, administered for 21 days continuous model.; With glomerular apoptosis index than serum creatinine assessed adenine fed rat model of chronic renal failure, kidney function more reliable. |
