Objective: To investigate intervention of LiCl on aggressive behavior of social-isolated golden hamsters through praxiology test and compare and the modify of neurotransmitter.Method: 70 male hamsters were randonmly divided into 6 groups on postnatal day 29(P-29). Normal group(NG),positive group(PG),LiCl low group,middle group and high group (LG,MG,HG) were all 10, intruder group were 20 (To test aggressive of other groups, not used in the end-result). Food and water were available throughout the experiment. The animal were under a reverse light/dark cycle (14L:10D, lights off at 9:00am). NG and intruder group were communal, 5 in one cage. Other group individually housed. LG,MG and HG were administered with 25,75,225mgLiCl/kg·weight in distilled water once a day after P-39, NG were gaven 1ml distilled water at the same time. Hang test were carried out for all the animal on P-72,P-73,P-84 once a day to measure muscle strength. Open-field test were carried out to evaluate reactiveness level and curious to fresh environmental. Beginning on P-76, animals were repeatedly tested aggressive for 5 consecutive days, and intruders were used to faver responses of other groups. All the animals did not administered with LiCl solution or distilled water on P-81, and weigh them. Get blood from their heart and put them to death. Get brain and testicle to measure organ index. Brain were made to 10% tissue homogenate to test the content of 5-HT,5-HIAA,DA and the activity of NOS,MAO, and test the .content of 5-HT,5-HIAA and DA of serum.Result: Weight increase of PG was less then NG (P<0.05), but there is no significant difference between LG and NG (P>0.05), HG was less then PG and LG (P<0.05); testicle index of PG was larger than NG (P<0.01), but their was no significant difference between 3 groups of LiCl and NG (P>0.05), LG and HG was less then PG (P<0.01, P<0.05); their was no significant difference between PG and NG on brain index (P>0.05), but HG was larger then PG and NG (P<0.01), and larger then NG and MG (P<0.05); the aggressive behavior score of PG was larger then NG (P<0.01), there was no significant difference between LG and PG (P>0.05), MG was less than other groups (P<0.01), HG was less than PG and LG (P<0.01, P<0.05); there was no significant difference among all the groups on crossing (P>0.05).PG was lager than NG on rearing(P<0.01), all LiCl groups were less than PG (P<0.01); there is no significant difference between PG and NG on hang time (P>0.05), all LiCl groups were less than PG (P<0.01); content of 5-HT in brain of PG was less than NG (P<0.05), there was no significant difference between MG,HG and NG (P>0.05), and MG was larger than PG (P<0.05), the relationship between content of 5-HT and aggressive behavior score was negative correlation (r=﹣0.347,P<0.05); content of 5-HIAA in brain of PG was less than NG (P<0.01), there was no significant difference between all the Licl groups and PG (P>0.05), but showed a upward trend, the relationship between content of 5-HIAA in brain and aggressive behavior score was negative correlation(r=﹣0.553, P<0.01); content of 5-HIAA in serum of PG was less than NG (P<0.01), there was no significant difference between all the MG,HG and NG (P>0.05), but MG,HG were larger than PG (P<0.05), the relationship between content of 5-HIAA in serum and aggressive behavior score was negative correlation (r=﹣0.317, P<0.05); there was no significant difference between PG and NG on DA in brain (P>0.05), there was no significant difference between all LiCl groups and PG(P>0.05), but there were a upward trend, the relationship between content of DA in brain and aggressive behavior score was positive correlation (r=0.393, P<0.01); content of 5-HT and DA in serum and activity of NOS and MAO in brain were no significant difference between all the groups, these indexes had no correlation with aggressive behavior score.Conclusion: It was feasible to to establish aggressive model of golden hamsters in experiment. Hamsters were aggressive through social-isolated. The aggressive behavior induced by social-isolated was significantly depressed by given 75mgLiCl/kg once a day for 42 days. The mechanism of it maybe concern with the elevated content of 5-HT of brain and 5-HIAA in serum.
