Both prokaryotic bacteria and eukaryotic human being, when exposing to high temperature or other stress factors, will synthesize a group of proteins called heat stress proteins ( HSPs). HSPs can make cells or organisms recover from different kinds of stresses and protect them from being impaired. HSPs, function as molecular chaperones, are highly conserved proteins during evolution, its polymorphism could be associated with either susceptibility to, or protect from, diseases or conditions secondary to various types of stresses imposed on cells or organs. Among them, HSP70 family are most important and a lot of researches have been done about HSP70. HSP70-1, HSP70-2 and HSP70-hom gene have been located in chromosome 6p21.3, the MHC III region, and may be associated with some kinds of diseases.Although there have been lot of researches about the association of HSP70 polymorphism with different diseases, but the relationship between HSP70 polymorphism and altitude stress caused by hypoxia haven't been reported.To investigate the relationship between HSP70 polymorphism andaltitude stress, in present study, we chose 72 soldiers with altitude stress and 173 soldiers as control. HSP70-1, HSP70-2 and HSP70-hom genotypes were analyzed by the polymerase chain reaction- restriction fragment length polymorphism(PCR-RFLP) technique.The results showed that the HSP70-1 polymorphism was similar in two groups. In contrast, the genotype frequencies of HSP70-2 and HSP70-hom were significantly different in the altitude stress group than in the control. HSP70-2 B/B genotype and HSP70-hom A/A genotype frequency were significantly increased in the altitude stress group (p<0.05) .The results suggested there was a significant association of HSP70-2 B/B genotype and HSP70-hom A/A genotype with altitude stress. Maybe individuals with the genotype had weaker adaptive ability than the others, this finding may help us find the high susceptibility group in professional groups and protect the health of these people.Low temperature is one of the ill weather conditions of physical factors, long-term work under low temperature can do harm to cardiac system, immune system, CNS, bone and joint system. But a little research has been done on cold stress in labour hygiene field. With the development of economy, more and more people are working under low temperature, such as cold or freezing storage. It's worthy of finding if the HSP70 level and the titre of its antibody changed respectively.We chose 53 workers who work under 0℃ and 53 workers whowork under -20 ℃ as our research group, and 58 villagers as control. Using Western-dot-blotting, the level of HSP70 was determined. The titre of antibody to HSP70 in plasma was determined by Western blot?ELISA . The activities of SOD and NOS, and the concentration of GSH, MDA, NO in plasma of the soldiers of different group were determined with kits.The results showed that HSP70 levels increased significantly in both 0℃ and -20 ℃ group than in control group (p<0. 05), and there was no difference of HSP70 between 0℃ and -20 ℃ group (/?>0.05 ) . As to the antibody, there were 3 positive ones with 1:10 (positive rate was 5%) in control and 14 positive ones in 0℃ group(positive rate was 26%), 11 positive ones in -20 ℃ group (positive rate was 21%). Among these, there was 1 positive case with 1:20 (positive rate was 2%) in control group and 8 positive ones in 0℃ and -20 ℃ group (positive rate was 15%) respectively. There was a significantly increase of antibody to HSP70inO℃ and -20 ℃ group than in control (p<0.05) . In addition, there was 1 positive case with 1 :40(positive rate was 2%) in both 0℃ and -20 ℃ group, but no case with 1:40 positive was found in control group. There was no significantly difference of antibody level between 0 ℃ and-20℃ group.The levels of SOD, GSH were lower in 0℃ and -20 ℃ group, but MDA, NO and NOS were higher in O'C and -20℃ group than in the control (p<0.05 ) .These results suggested that the levels of HSP70 in 0℃ an...
