Study The Tissue Distribution, Survival And The Effect To Important Organ Of Paclitaxel Drugs On Nude Mice With Ovarian Cancer In Situ Tansplantation

Objective：Study the tissue distribution, survival and the effect to important organ of taxoldrug, in ovarian cancer orthotopic transplantation nude mice, for better clinicalapplication of paclitaxel drug in treatment of ovarian carcinoma and further study ofits mechanism, providing a scientific basis.Methods：1. Establishment of orthotopic mouse model of human ovarian carcinoma tumorof ovarian cancer tissue block will be cut into about1~2mm3sizes, inoculated in8~10-week-female-nude-mice ovary, built the experiments required models.2.21model mice are randomly divided into3groups: Paclitaxel liposome group(3dose groups, the dosage is10mg/kg,15mg/kg,30mg/kg, n=3) Paclitaxel injectiongroup (3dose groups, the dosage is10mg/kg,15mg/kg,30mg/kg, n=3),,5%glucosecontrol group (n=3) The equal volume of the liquid at the corresponding density areinjected to the model mice by tail vein at the11th13th15thdays. The groups arecompared in survival situation and effection to main organ.3. Take24models, randomly divided into2groups: Paclitaxel liposome groupand Paclitaxel Injection group.inject the drug at the concentration of15mg/kg at the21th day. After the administration of1hours,8hours,15hours,30hours, kill3nudemice from each group randomly. Take liver tissue and metastases, splenic tissue andmetastatic tumors, ovarian tumor tissues and peripheral blood,and detect the drugconcentrationof them by high performance liquid chromatographic (HPLC).Results:15mg/kg group of paclitaxel liposome’s mean maximum survival time is5.33±1.53days, life extension rate is100%.30mg/kg group of paclitaxel injection’saverage survival time for a minimum is1.67±1.16days, life extension rate is-37.45%.Group15mg/kg of the same drug groups get the maximum survival time.The samedoses of two drugs, taxol liposome get longer survival time.HPLC show theresults:the drug distribution difference of paclitaxel liposome and Paclitaxel injection has no statistical significance (P>0.05) in ovarian and ovarian tumor tissues; in theliver and spleen and metastatic tumor tissue, paclitaxel liposome’s distribution ismore than Paclitaxel injection,and has statistical significance (P＜0.05). Pathologicalexamination, paclitaxel liposome give more influence than Paclitaxel injection groupin liver and spleen, give less effects to the cardiorespiratory than Paclitaxel injectiongroup, two groups of drugs has no effect to kidney basically.Conclusion:Paclitaxel liposome could improve the survival time of human ovariancarcinoma orthotopic transplantation nude mice, the same dose of PaclitaxelLiposomal has more advantages.Liposomal paclitaxel in metastatic ovarian tumortissues with higher concen-tration and longer lasting time, stronger tumor suppressoreffect. smaller effection to the heart, lung and other important organs.
