Objective:To investigate the effectiveness of administered S-100 anti-adhesion solution in preventing postoperative intraperitoneal adhesion formation and its optimal concentration interval and its effect on the level of transforming growth factor betal in serum.Methods:Sixty Wistar rats were divided into six groups at random:model control group,sodium hyaluronate group,0.5% concentrationS-100group,3% concentration S-100 group,5% concentration S-100 group and TISTAT(?) group, each containing ten rats. The animals underwent a laparotomy and adhesions were induced to establish the postoperative abdominal adhesion model. Befor the abdominal wall was closed, the operation region were smeared with different material in every group:the model control group with 2ml physiological saline, the sodium hyaluronate group with 2ml sodium hyaluronate, the 0.5% concentration S-100 group with 2ml 0.5%S-100, the 3%concentration S-100 group with 2ml 3%S-100, the 5%concentration S-100 group with 2ml 5%S-100 and the TISTAT(?) group with TISTAT(?) absorbable hemostatic gauze. Second-look operations were performed 10 days later, we conducted the adhesion scores gradation judging,hematoxylin and eosin stain,immunohistochemisty and Hydroxyproline assay to observe the effect of S-100 anti-adhesion solution and evaluated its influence on the level of transforming growth factor betal in serum. Results:1. By the results of the adhesion scores gradation judging,hematoxylin and eosin stain and Hydroxyproline assay, there was statistically significant difference between the sodium hyaluronate group,0.5% concentration S-100group,3% concentration S-100 group,5% concentration S-100 group and TISTAT(?) group with model control group(P<0.05); There was no statistically significant difference between the sodium hyaluronate group with 0.5% concentration S-100 group and no statistically significant difference between the 3% concentration S-100 group,5% concentration S-100 group and TISTAT(?) group by paired comparison (P>0.05); The comparison between any group of the sodium hyaluronate group and 0.5% concentration S-100 group with any one of the 3% concentration S-100 group,5% concentration S-100 group and TISTAT(?) group showed statistically significant difference in adhesion formation (P<0.05).This outcome indicated that sodium hyaluronate,S-100 anti-adhesion solution and TISTAT(?) all can significantly reduce the incidence, severity and extent of the postoperative peritoneal adhesionin formation and the 3% concentration S-100,5% concentration S-100 and TISTAT(?) had better effect than the sodium hyaluronate and 0.5% concentration S-100.2. Immunohistochemical study results showed that the adhesion tissue Collagen I content in the sodium hyaluronate group,0.5% concentration S-100group,3%concentration S-100 group,5% concentration S-100 group and TISTAT(?) group was lower than model control group and had statistically significant difference (χ2=11.098, P=0.049<0.05), this indicated that Sodium Hyaluronate,S-100 anti-adhesion solution and TISTAT(?) had the efficacy of reducing the content of adhesion tissue CollagenⅠ; By paired comparison there was no statistical significant difference between sodium hyaluronate,S-100 anti-adhesion solution and TISTAT(?), this indicated that their effects had no difference. 3. By the measurement results of serum transforming growth factor betal, sodium hyaluronate,S-100 anti-adhesion solution and TISTAT(?) were not statistically significant compared with model control group although their levels were lower than model control group (F=0.161, P=0.976), this did not prove that they can effect the serum transforming growth factor betal level.Conclions:S-100 anti-adhesion solution were effective in prevention of peritoneal adhesion formation in a rat model and its efficacy was better than sodium hyaluronate, its best concentration interval was from 3% to 5%.
