Objective:To study the expression and the role of nuclear factor peroxisome proliferator activated receptor-Y(PPAR-y) and Cyclooxygena-se-2(COX-2) in placenta of patients with pre-eclampsia.Methods:30 cases of normal pregnant women (control group) and 57 women with pre-eclampsia (25 cases of mild pre-eclampsia group; 32 cases of severe pre-eclampsia group.) were selected. The expression level of PPAR-y and COX-2 in placenta among the two groups was determined by immunohistochemistry.All cases had no primary hypertension,nephritis,diabetes or other diseases.There were no significant difference in the age, gestational weeks and body mass index in the two groups,so date of the two groups had comparability.Results:l.The expression of PPAR-y in placental villus PPAR-y mainly expressed in the nucleus of trophoblast cells in placental villi. Compared with the control group, the expression level of PPAR-y in patients with pre-eclampsia significantly increased and the difference was statistically significant(p<0.05). The expression level in patients with severe pre-eclampsia was significantly higher than in patients with mild pre-eclampsia and the difference was statistically significant(p<0.05).2.The expression of COX-2 in placental villus COX-2 mainly expressed in the cytoplasm of trophoblast cells in placental villi. Compared with the control group, the expression level of COX-2 in patients with pre-eclampsia significantly increased and the difference was statistically significant(p<0.05). The expression level in patients with severe pre-eclampsia was significantly higher than in patients with mild pre-eclampsia and the difference was statistically significant(p<0.05).3.Correlation analysis showed that the expression of PPAR-γand COX-2 in placent of patients with pre-eclampsia had a positive correlation.. Conclusions:The high levels of PPAR-γand COX-2 in placenta of patients with pre-eclampsia may be related to the etiology of pre-eclampsia.
