Objective: Studyding the expression of the aggrecanases ADAMTS-5 in human intervertebral disc tissue, and exploring the influence of ADAMTS-5 in the process of Intervertebral disc degenerativeMethod: The experimental group consisted of 30 cases (male 15 , female 15) of the fresh pulpy nucleus tissues which were obtained from patients with lumbar disc heriation by surgical intervention in the period of march 2009 to september 2009 , the mean age is 40.58 (24-63).28 cases (male 18 , female 10) of the fresh pulpy nucleus tissue harvested by operation for fracture of lumbar vertebrae acted as control group , the mean age is 15.25(13-22) . the protein expression of ADAMTS-5 were detected with haematoxylin-eosin and Immunohistochemistry.Result: Hematoxylin and eosin staining revealed obvious signs of degeneration in Degenerated sections.in the pulpy nucleus tissues the characteristic were changed , the cell quantity reduce, lose the original cartilage cell shape. Use Immunityhistochemistry to examing the ADAMTS-5 protein expression , we found , in the control group, the most sample ADAMTS-5 masculine cell rate is low, The experimental group , the most sample ADAMTS-5 masculine gender rate is high . Using the x2test analysis confirmation groups the difference has statistics significance P(<0.05).Conclusion: The aggrecanases ADAMTS-5 may contribute to the changes occurring in the ECM during intervertebral disc degeneration. Targeting these enzymes may be a possible future therapeutic strategy for the prevention of intervertebral disc degeneration and its associated morbidity.
