Glioblastoma is one of the most common primary malignant tumors in central nervous system. It can't be completely removed by operation and is easy to relapse. However, it is moderately sensitive to radiotherapy and chemotherapy. Therefore tumors can't be radical cured by the sequential therapy: surgery combined with radio-chemotherapy, which is the available standardized international accepted treatment protocol nowadays.Bio-immunotherapy is a kind of treatment mainly using biological engineering principal to arouse patients'autoimmune function to improve life quality, inhibit tumor recurrence and enhance therapeutic effect by means of antibodies therapy, cytokines therapy, adoptive immunotherapy, gene therapy, etc. It is the fourth treatment mood after-present of surgery, radiotherapy and chemotherapy. Compared with the latter three, Bio- immunotherapy has its advantages that were more safety, more effective and lower side effect.This topic is aim to observe the biologic activity of the tumor-cell vaccine which has been prepared by heat-shock mean to allergize human peripheral blood-derived DCs with the glioblastoma. from a glioblast specimen by intra operation frozen section to load with peripheral blood-derived DCs after the apoptosis of glioblastoma induced by heat-shock. To investigate the safety and effectiveness of such vaccine that has been prepared and performed lymphonodes infusion.Partâ… Test of Dendritic Cells Loaded with Apoptosis of Glioblastoma Induced by Heat-shock and Activated T cells In vitro.Objective: To prepare dendritic cell (DC) loaded with apoptosis of glioblastoma induced by heat-shock. Discuss its'activity against the tumor.Methods: Induced human peripheral blood-derived immature DC. DC loaded with apoptosis of glioblastoma induced by heat-shock. Detect its phenotype by Fluorescence double labeling flow cytometry and test the activity of cytotoxic T lymphocyte (CTL) by MTT colorimetric assay.Results: DC had fine activable effect on CTL. As far as the function of killing glioblastoma, CTL activated by DC loaded with heat-shock glioblastoma were significantly higher than DC loaded with freeze-thaw-cracking glioblastoma, with a higher effector-target ratio resulting in higher killing.Conclusion: DCs loaded with heat-shock glioblastoma has its function of killing glioblastoma through active process on CTL. Prepare the vaccine which is coincidence with autologous Glioblastoma-DC and apply for clinical trial.Methods : Prepare the vaccine which is coincidence with autologous Glioblastoma-DC from a glioblast specimen by intra operation frozen section to load with peripheral blood-derived DCs after the apoptosis of glioblastoma induced by heat-shock. The project to prepare such vaccine for clinical trial accordance with clinical application is under the ratification of ethical committee of hospital. Set up control group with the treatment of traditional surgery and radio-chemotherapy post-operation, while therapeutic group combining with the application of Glioblastoma-DC vaccine and the radio-chemotherapy. Compare the immune functional parameters: tumor control, rate live time and Karnofsky scores and the complication between the two groups. Discuss the safety and effectiveness for glioblastoma treatment.Results: After clinical therapy and follow-up investigation, statistics shows that the three immune functional parameters: tumor control, rate live time and Karnofsky scores are better in therapeutic group than in control group.Conclusion: Combining with the application of Autologous Glioblastoma-DC vaccine and the radio-chemotherapy has a certain function of anti-tumor in vivo. Preliminary clinical trial shows its satisfactory safety. That offers a more effective conception for glioblastoma treatment.
