Objective: To detect the expression of Talin1 protein and mRNA in the normal liver tissues,HCC tissues and paracarcinomatous tissues and investigate the relationship between Talin1 expression and the oncogenesis and prognosis of HCC.Methods: The expression of Talin1 protein was detected by immunohistochemical SP method in the normal liver tissues(n=10),HCC tissues(n=32) and its paracarcinomatous tissues(n=32).The expression of Talin1 mRNA was also determined by Real Time reverse transcriptase PCR in the three tissues.Results: The expression of Talin1 protein was observed in the normal liver tissues,HCC tissues and paracarcinomatous tissues.The positive rate of Talin1 protein were 100%,62.5%,96.9%respectively.The brown granules of positive cell were mainly distributed in the cytoplasm and cell membrane.The expression of Talin1 protein in HCC tissues was significantly lower than that in the paracarcinomatous tissues and normal liver tissues(P<0.05),but not between paracarcinomatous tissues and normal liver tissues(P>0.05).Talin1 protein expression were not related to sex,age,tumor number,the expression of AFP and HBsAg(P>0.05),and has no correlation with the One-year disease-free survival of patients(P>0.05),but were significantly correlated with pathological differentiation of HCC,The integrity of tumor capsule,Portal vein tumor thrombus and tumor size(P<0.05). The expression of Talin1 mRNA in HCC tissues and paracarcinomatous tissues was significantly lower than that in the normal liver tissues(F=24.343,P<0.01),but not between HCC tissues and paracarcinomatous tissues(P>0.05). There was no significant difference between expression of Talin1 mRNA and sex,age,the expression of AFP and HBsAg(P>0.05),The expression of Talin1 mRNA in HCC tissues has no correlation with the One-year disease-free survival of patients(P>0.05),but were significantly correlated with pathological differentiation of HCC,The integrity of tumor capsule,Portal vein tumor thrombus,tumor number and tumor size(P<0.05).Conclusion: Talin1 is related to the carcinogenesis,infiltration and metastasis of HCC;The level of Talin1 expression might be a valuable marker for diagnosis and prognosis of HCC.
