OBJECTIVE: To study the significance of protein expression of tumor suppressor gene PLAGL1 in humor hepatcellular carcinoma(HCC) and mRNA expression in L-02 and HepG2 cell line.METHODS: 30 cases of HCC tissue and 30 cases of tumor-adjacent tissues were collected from patients surgically treated at our hospital from June to August 2009. The protein expression of PLAGL1 was detected by immunohistochemistry, and its significance was analyzed. PLAGL1 mRNA expression level was detected by RT-PCR in both normal liver cell line L-02 and hepatoma cell line HepG2 cells.RESULT: PLAGL1 protein expression in tumor-adjacent tissues(n=27, 90%) was absolutely stronger than hepatocellular carcinoma tissues(n=2, 7%) (χ2=38.44, P<0.05). RT-PCR results showed that PLAGL1 mRNA in the L-02 cells was significantly higher than HepG2 cells.CONCLUSION: PLAGL1 protein expression was parallelled with the PLAGL1 mRNA downregulation. Downregulation of PLAGL1 protein may be correlated to the occurrence and development of HCC.
