Objective To explore the expression of tumor suppressor gene RUNX3 protein in gastric cancer, colorectal cancer tissues, and comparative analysis of both the relevance and clinical significance. To further understand the tumor suppressor gene RUNX3's role in the digestive tract, for their theoretical research, clinical diagnosis and treatment help.Methods Collected 52 cases of resected gastric cancer patients and the same period 41 patients with rectal cancer surgical specimens, to study which tumor suppressor gene RUNX3 protein in gastric cancer, rectal carcinoma study:① Gastric cancer, colorectal cancer organizations and the corresponding adjacent normal tissue morphology change were observed by HE staining;②Tumor suppressor gene RUNX3 protein in gastric cancer, colorectal cancer tissues and clinical correlation were detected by immunohistochemical SP method;③Connection of RUNX3 protein in gastric cancer, colorectal cancer tissue were contacted by computer image analysis;④UNX3 protein in gastric cancer, rectal carcinoma were detected by Western blot;⑤Immunohistochemical analysis of data mean optical density value, and Western blot gray value were processed by Statistical software SPSS for Windows13.0 image.Results①HE staining showed normal tissue adjacent to gastric cancer, rectal wall from outside to the inside of four tiers, mucous layer, submucosa, muscle, membrane, mucous membrane layer was divided into epithelium, lamina propria, muscularis mucosa. HE staining showed histological type adenocarcinoma mainly common with tubular adenocarcinoma in gastric cancer; carcinoma of rectum histological type, the well-differentiated tubular adenocarcinoma and papillary adenocarcinoma more common.②Immunohistochemical staining showed that 52 cases of gastric cancer of corresponding adjacent normal tissues could be expression of RUNX3 protein, mainly located in the surface mucous cells and gastric glands in the cytoplasm; 32 cases (61.5%) RUNX3 protein expression in 52 cases of gastric carcinoma and the lower level of differentiation, the less positive expression, and there was coloring the cytoplasm and nuclei of heterogeneity; 38 cases (92.7%) in 41 cases of rectal mucosa adjacent to the corresponding normal tissue expression of RUNX3 protein, located in the cytoplasm of columnar epithelial cells,19 cases (46.3%) in 41 cases of rectal cancer tissues were positive, which lower than the positive rate of RUNX3 protein in gastric cancer (P<0.05); Immunohistochemical image analysis showed OD values that RUNX3 protein expression in gastric cancer group was better than in colorectal cancer group, the difference was significant (P<0.05);③Western blot results showed that 38 cases (73.1%) in 52 cases of gastric carcinoma reduced or even missed the RUNX3 protein expression, the RUNX3 protein expression in normal gastric mucosa was significantly higher than the corresponding poorly differentiated gastric cancer tissue (P<0.05); RUNX3 protein in 15 cases (36.6%) in 41 cases of rectal cancer tissue were positive, RUNX3 protein expression in normal colonic mucosa was significantly higher than the poorly differentiated corresponding colorectal tumor tissue(P<0. 05).Conclusions①HE staining shows that gastric cancer, rectal cancer, the corresponding adjacent normal tissue (from cancer tissue≥10 cm) can be used as normal control group; type of gastric adenocarcinoma mainly common with tubular adenocarcinoma; rectal cancer to be in well-differentiated tubular adenocarcinoma and papillary adenocarcinoma more common.②RUNX3 protein in gastric cancer tissues compare with positive expression rate of normal gastric mucosa significantly reduced, which shows that decreased expression of RUNX3 protein in gastric cancer may be related to the occurrence, development.③RUNX3 protein in tumor tissue with positive expression rate in the adjacent mucosa significantly reduced, which shows that decreased expression of RUNX3 protein in colorectal cancer may be related to the occurrence, development.④RUNX3 protein expression in human gastric cancer group is higher than OD values of colorectal cancer group, RUNX3 protein expression down-regulation is likely more meaningful in the diagnosis of gastric cancer than the carcinoma of rectum.⑤RUNX3 expression and gastric cancer patients has no associated with sex, age, tumor location, tumor invasion depth, size, Lauren classification, but has associated with the differentiation degree and lymph node metastasis; RUNX3 protein expression in rectal cancer patients has no associated with sex, age, tumor location, size, but has associated with the degree of differentiation, tumor depth of invasion, Dukes stage and lymph node metastasis.⑥To explore in depth the structure and function of Runx3 gene expression regulation mechanisms and regulatory network in gastro-intestinal tumorigenesis role in the process, for the diagnosis and treatment of gastrointestinal tumors and provide a new biological markers and tumor gene therapy of the target from the morphology, molecular level.
