Objective To detect the expression of FHIT gene protein and elucidate thepotential relationship between FHIT protein expression and clincopathlogical indices in human colorectal carcinoma.Methods FHIT gene protein expression was detected in 90 cases of formalin-fixed,paraffin-embedded cancerous and matched adjacent normal colorectal specimens by S-P immunohistochemical method and FHIT gene protein expression was detected in cancerous and matched adjacent normal colorectal tissue of 30 patients with colorectal carcinoma by Western Blotting. The relations between FHIT protein expression and clincopathlogical indices such as gender, age, tumor site, tumor size, histologic type, invasive depth, differentiation, Dukes stage and lymph nodes metastases was analyzed.Results The results of immunohistochemistry display that the positive ratio ofFHIT gene protein expression in colorectal carcinomas(47.8%, 43/90) was significantly lower than that in matched adjacent normal colorectal mucosa (100%, 90/90) (P<0.05), the positive ratio of FHIT gene protein expression in carcinoma with serosa infiltration(23.3%,7/30) was significantly lower than in carcinoma without serosa infiltration (60.0%,36/60) (P<0.05), the positive ratio of FHIT gene protein expression in high differentiation(61.5%,24/39), moderate differentiation (48.1%,13/27) and Low differentiation (25.0%,6/24) was significantly different(P <0.05), the positive ratio of FHIT gene protein expression in carcinoma of Dukes C and D stage...
