Expression and Clinincal Significance of RECK and MMP-9 in Human CholangiocarcinomaOBJECTIVE:To investigate the expression of thereversion inducing cysteine-rich protein with Kazalmotif (RECK) and Membrane-type matrix metalloproteinase-1 (MT1-MMP/MMP-9) in human cholangiocarcinoma tissues;Para-tumor tissue and normal bile duct tissues.Investigate the expression of RECK and MMP-9 in cholangiocarcinoma,reveal the relationship of them in cholangiocarcinoma and whether existing correlation between of them.METHODS:The 60 specimens of cholangiocarcinoma were selected from January,2003 to December,2008 at the People's Hospital of Sanxi province ,and all pathologically validated. no tumor cell were found pathologically in Para-tumor tissue and 18 case of normal bile duct tissues. We detected the exp-ression of RECK and MMP-9 in the tumor and Para-tumor tissue in immunologichistochemistry methods,and utilized the image analysis software (Image-pro Plus 5.0) to count and analysis the specimens.Statistic analysis was completed with statistic software SPSS 11.0,and K-S test,analysis of variance,SNK test and t test were used.The analysis of correlation of each factor used linear correlation.P<0.05,was as the level for statistical significance.RESULTS:RECK and MMP-9 were mainly expressed on cytoplasm.The expression of MMP-9 was significantly higher in cholangiocarcinoma tissue than in para-tumor tissue (p<0.05);But expression of RECK significantly higher in para-tumor tissue than in cholangiocarcinoma tissue (p<0.05).Further study indicated the expression of RECK and MMP-9 had close relationship with TNM classification (p<0.05).There is a negative correlation is between RECK and MMP-9.CONCLUSIONS:The expression of MMP-9 in the tumor tissue was higher than that in the para-tumor tissue,the expression levels of MMP-9 upregulate obviously along with the progression of tumor pathological stage.But the expression of RECK in the tumor tissue was lower than that in the para-tumor tissue, the expression levels of RECK descends obviously along with the progression of tumor pathological stage.The expression of RECK and MMP-9 were highly correlated with the pathologic degree of tumor. RECK and MMP-9 might play an important role in the pathogenesis of cholangiocarcinoma.RECK and MMP-9 could be considered as the marker for predicting the metastasis and recurrence of cholangiocarcinoma.
