Objective:1.Determination of benign and malignant lesions of carcinoma andadjacent normal tissue RECK/MMP-9expression of both the expression ofdifferences in different tissues.2.Analysis of RECK/MMP-9expression correla-tion between the level of the biological behavior of breast cancer and theclinical and pathological data.3.Explore the mutual association of RECK/MMP-9expression in breast cancer tissue and joint detection value,and provide abasis for future related to the development of anticancer drugs.4.Preliminaryanalysis of RECK expression and the prognosis of breast cancer patients.Method: Select Mianyang Central Hospital clinical and pathological dataintegrity in2004-2007,78cases of paraffin-embedded specimens of breastcancer tissues and adjacent normal tissue,40cases of benign breast lesionstissue.Using immunohistochemistry (S-P) were determined of RECK/MMP-9protein expression in these three organizations. A retrospective analysis of therelationship between the RECK/MMP-9protein with clinical pathologicalparameters information,including age, tumor size, histological grade,ipsilateralaxillary lymph node metastasis, estrogen receptor (ER) and progesteronebybody (PR),the proto-oncogene (Her-2), cell proliferation factor Ki-67Rate,all the results,using SPSS17.0for windows statistical packagehandling.Postoperative follow-up data of56cases of breast cancer patientsusing the Kaplan-Meier method of survival analysis test,overall survival usinglog-rank test on the follow-up period of RECK positive and negative groups(Overall survival, OS), the impact of significant with analysis,multivariate analysis using Cox hazards regression model was completed.Result:1.RECK in breast cancer tissue, benign lesions and adjacent normaltissues, the positive expression rate of48.72%(38/78),67.50%(27/40),87.18%(68/78),was gradually increasedtrend,(P <0.05).2.MMP-9in breast cancertissue,benign lesions and adjacent normal tissues, the positive expression rateswere67.95%(53/78),32.50%(13/40),28.12%(22/78),was gradually downwardtrend,the difference was statistically significant (P <0.05).3.breast cancerRECK expression and age, tumor size, menopausal conditions, the ER, PR, andKi-67was no significant correlation (P>0.05),with histological grade, clinicalgrade, axillary lymph node metastasis, Her-2was significantly (P <0.05).Theexpression rate with the histological grade, clinical stage increased graduallydecreased (P <0.01).4.breast cancer tissue MMP-9and age, menopausalconditions,PR and Ki-67no significant correlation (P>0.05),and tumorsize,histological grade,clinical stage, axillary lymph node metastasis, ER, Her-2significantly correlated (P <0.05). The expression rate with the histologicalgrade, clinical stage higher and higher (P <0.01).5breast cancer, RECK andMMP-9expression was negatively correlate-d (r=-0.541, P <0.01).65-year follow-up data of56cases of breast cancerbreast cancer patients tumor-free survival (Disease Free Survival Rate83.39%,the DFS), the OS94.64%. Log-Rank test showed that RECK expressionnegative group and positive group of follow-up period the survival curves, thereare significant differences of RECK positive were followed up for overallsurvival was significantly higher than the negative group(P <0.05). Histologicalgrade III, Her-2(+) and ER (-) expression of positive affect the survival time ofpatients with breast cancer risk factors. RECK expression status is anindependent prognostic indicator in breast cancer patients (P=2.655,<0.05).Conclusion:1.RECK in breast cancer was significantly lower than the benignlesions and adjacent normal tissue.RECK expression and clinical stage, histological grade, lymph node metastases and Her-2expression was negativelycorrelated. Prompt with the increased degree of malignancy of breast tissue,expression of RECK decreased.2.MMP-9in breast cancer tissues was higherthan that of benign breast lesions and adjacent normal tissue. MMP-9expression and breast cancer tumor size, clinical stage, histological grade,lymph node metastasis, ER and Her-2expression was positively correlated. TipMMP-9may be involved in breast cancer development and transferprocess.3.breast cancer, RECK and MMP-9expression was negatively correlat-ed,of RECK loss of expression of MMP-9overexpression may be the malignanttransformation of breast tissue and breast cancer are invasion, metastasis ofimportant biological indicators. The joint test of both breast cancer developme-ent and prognosis of great significance.4.56cases of breast cancer patients ofRECK-positive group than negative group,5-year survival rate. Histologicalgrade III, Her-2(+) and ER (-) affect the survival time of patients with breastcancer risk factors. RECK is an independent risk factor for breast cancerprognosis.
