Objective:To detect the expression of Her-2/neu and NF-κBp65 in gastric carcinoma tissue, to explore their relationship and the role in gastric cancer formation, and the relationship with clinicopathologic characteristic in gastric carcinoma.Method:To collect 80 cases of gastric carcinoma and 30 cases of normal gastric tissue in the first hospital affiliated of Shan Xi Medical University from 2007.1 to 2008.12. To detect the expression of Her-2/neu and NF-κBp65 in gastric carcinoma by immunohistochemistry assay (PV-9000) and to analyze the relationship with clinicopathologic characteristic in gastric carcinoma.Result:The positive rate of Her-2/neu expression in gastric carcinoma tissue was 22.5% (18/80) and in normal tissue was 3.3%(1/30), there is significant difference between them (P<0.05). There was no significant difference among the expression of Her-2/neu in different sex,ages,tumor size,location,invasion depth,metastasis and TNM stages of the cancer, while associate with degree of histodifferentiation and lymphatic metastasis (P<0.05). The positive rate of NF-κBp65 expression in gastric carcinoma tissue was 61.25%(49/80) and in normal tissue was 26.7%(8/30), there is significant difference between them (P<0.05). There was no significant difference among the expression of NF-icBp65 in different sex,ages,tumor size,location,metastasis and TNM stages of the cancer, while associate with degree of histodifferentiation,invasion depth and lymphatic metastasis(P<0.05). Her-2/neu is positively correlated with NF-KBp65 in gastric cancer tissue (r=0.244, P<0.05).Conclusion:Her-2/neu and NF-κBp65 were highly expressed in gastric carcinoma tissue and were associated with the occurrence and development of gastric carcinoma, they can be used as the ideal targets for molecular biology treatment of gastric carcinoma.
