Objective:To investigate the expression of hTERC gene, HPV 16/18,31/33 DNA, Rb-related tumorgene proteins P16ink4a,CyclinD1,CDK4,Rb,E2F-1 and Ki-67 in invasive squamous cell carcinoma of cervix(ISCC) and squamous intraepithelial lesion (SIL). To analyze their correlation to clinical pathological parameters of ISCC. So as to provide the scientific evidence for the early clinical diagnosis, genetic therapy and tureover predict of ISCC.Methods:73 cases of ISCC,30 cases of high-grade squamous intraepithelial lesions (HSIL),32 cases of low grade squamous intraepithelial lesion (LSIL), and 21 cases of normal cervical squamous epithelium (NCE) were collectted. hTERC gane was studied by fluorescence in situ hybridization (FISH). HPV DNA 16/18,31/33 was detecte by Chromogens in situ hybridization(CISH). P16ink4a, CyclinD1, CDK4, Rb, E2F-1 and Ki-67 proteins were studied by the tissue microarray and two-step immunohistochemistry. The results were analysised by SPSS13.0 version of statistical software.Rerults:(1)The frequency of hTERC gane in NCE,LSIL,HSIL,ISCC were separately 14.29%,47.26%,78.26%,77.78%(p<0.01). (2) The positive expression rate of HPV16/18 DNA in NCE,LSIL,HSIL,ISCC were 0,9.38%,46.67%and 65.75%. The expression of HPV16/18 DNA in the four groups was statistically significant different (p<0.01). Intravenous tumor thrombus more likely occur in HPV16/18 infection cases than non-infection (OR=3.875). (3) The positive expression rate of HPV31/33 DNA in NCE,LSIL,HSIL,ISCC were 0,12.50%,30.00%and 50.68%. The expression of HPV31/33 DNA in the four groups was statistically significant different (p<0.01). HPV31/33 infection sames relevant to the late stage (OR=3.5). (4) The expression of P16ink4a, CyclinD1, CDK4, Rb, E2F-1 and Ki-67 in the four groups was statistically significant different (p<0.01); Both Rb and Ki-67 positively correlated; With the expression rate of P16ink4a increasing, the malignancy degree of tumor cells decreased (OR=0.942); With the expression rate of CDK4 increasing, the malignancy degree of tumor cells increased (OR=1.033). (5) Elder with hige rate of clinical stage progress (OR=1.063)Conclusion:The amplification of hTERC gene can reflect severity of cervical squamous epithelial lesions and might be used as a marker predicting progression from LSIL to HSIL. HPV16/18,31/33 infection is closely related to the occurrence and development of cervical squamous cell carcinoma; HPV16/18,31/33 infection may result in hTERC gene amplification. CDK4, Ki-67 are valuable as an objective mark used in diagnosis of cervical squamous epithelial lesions; Rb gene expression in cervical squamous cell carcinoma need further research.
