Objective:To investigate the effect of propofol on rabbit liver during pneumoperitoneum.Methods:Twenty-eight rabbits were randomly divided into 4 groups (n=7):the normal control group (N group), the carbon dioxide pneum-operitoneum group (C group), the propofol group (P group) and the carbon dioxide pneumoperitoneum with propofol group (C+P group). Rabbits were anesthesiaed by 25% of the Ethyl Carbamate With 1.0g/kg BW by Auricular Vein. The pneumoperitoneum of C group and C+P group were established during 2hrs. The serums were collected at different time points (0,30,60, 90,120minutes after pneumoperitoneum) for detecting Alanine amino-transferase (ALT), aspartate aminotransferase (AST) and lactate dehyd-rogenase (LDH).2 hrs after pneumoperitoneum, rabbits were killed. Serums and tissue were collected. HE staining, morphological changes, Tumor necrosis factor (TNF-alpha), MDA malondialdehyde (MDA), superoxide dismutase Superoxide Dismutase (SOD), IL-6Interleukin-6 (IL-6) content and cytochrome P450 (CYP450) activity were measured.Results:1. Liver injury:the damage of C group and C+P group was observed at 60 minutes after carbon dioxide pneumoperitoneum. Howere, the damage of C+P group was significantly less than C group (P< 0.05). The levels of SOD, P450 in C group were significantly lower than C+P group (P<0.05), while the levels of MDA, TNF-a and IL-6 were significantly higher than C+P group (P<0.05). N group and P group remained relatively constant in almost all time.2. Two hours after pneumo- peritoneum, hepatocytes and sinusoidal endothelial cells were normal in both P and N group. C group showed significant expansion of sinusoidal, congestion, inflammatory cell infiltration in portal area, hepatic cells could be seen in varying degrees of cloudy swelling and adipose degeneration degeneration, spotty necrosis seen occasionally. C+P group showed obvious swelling of liver cells, sinusoidal dilation congestion, exchange controls is less obvious areas of inflammatory cells, liver cells had adipose degeneration, no significant necrosis.Conclusion:Propofol protects rabitts liver from carbon dioxide pneumoperitoneum.
