Extraction And Separation Of Chemical Components In Bean-Pot Of Mucuna Prurines(L.)DC.

Mucuna sempervirens, which is belonging to Leguminosae Mucuna family, can be used as Chinese traditional medicine as a whole plant. Some research showes that the seeds of Mucuna sempervirens contain a kind of drug named Levodopa, which is used for treating Parkinson's Disease. The cane has some medical function in activating blood and dissolving stasis and curing rheumatism and physical injuries. Although the whole plant has important medical value, there are not many articles involved the chemical constituents of Mucuna sempervirens. In order to investigate the components of Mucuna sempervirens, the chemical composition in its bean-pot was studied in this paper.The bean-pot was extracted with 95% ethanol and then concentrated. Degreased with petroleum ether, the aqueous solution was extracted with chloroform, ethyl acetate and n-butanol. The extracts of chloroform, ethyl acetate and n-butanol which were used for the further separation was obtained.One monomer in the chloroform extract was obtained by high-speed countercurrent chromatography (HSCCC) with a biphasic solvent system composed of TBME-mathanol-water (2:1:1, V/V) and a assisted use of preparative HPLC. Six monomers in the ethyl acetate extract and three monomers in the n-butanol extract were also obtained with a solvent system composed of petroleum-ethyl acetate-methanol-water (2:1:1: 1, v/v) and a solvent system composed of n-butanol-TBME-water (4:1: 5, V/V).The ten monomers were syringaresinol (â… ), vestitol (â…¡), formononetin (â…¢),2'-hydroxy-biochanin A (â…£), soybean isoflavone (â…¤), Sch 53823 (â…¥),3-hydroxy-2-methyl-2,3-dihydrobenzofuran-4-carboxylic acid (â…¦), 1-(4-hydroxybenzoyl)-glucose (â…§), kaempferol-3-O-glucorhamnoside (â…¨), kaempferol-3-O-rhamnogalactoside (â…«).
