The Effects Of Zhiganfang On Mitochondria Of Experimental NAFLD Rats

Objective:This study was designed to observe function disorder of mitochondria of NAFLD, the preventive effect of Zhiganfang on NAFLD and elucidate the mechanism。Methods:Twenty four Sprague-Dawley rats weighing 210±10g were randomly divided into four groups:8 for normal control group fed with regular chow,8 for model group fed with high lipid chow,8 for Zhiganfang-intervention group fed with high lipid chow, and the others for lipitor-intervention group fed with high lipid chow. Zhiganfang-intervention group rats were administered with Zhiganfang at dose of 12.5g/(kg.d)by intragastic infusion;lipitor-intervention group rats were administered with lipitor at dose of lmg/(kg. d)by intragastic infusion. All of the rats were killed at the end of sixteenth week. We take away all rats'livers, and calculate the liver index(liver wet weight/body weight×100%). The serous ALT, AST, TG,TC were assayed. The serous MDA and SOD were assayed. The ratio of serous SOD to MDA was calculated, too. The liver histopathology sections of HE staining were observed. UCP-2mRNA was detected by RT-PCR. The statistical analysis of the data was done with the statistical package of SPSS 13.0.Results:There is statistic significance of liver wet weight and liver index of the normal group compared with the other three groups (P<0.05); and there is no statistic significance among Zhiganfang-intervention group, Lipitor-intervention group and model group(P>0.05). There is statistic significance of AST and ALT between model group and normal group (P<0.05);and the as Zhiganfang-intervention group and model group(P<0.05). There is statistic significance of AST and ALT between Zhiganfang-intervention group and Lipitor-intervention group(P<0.05). There is statistic significance of TG, TC, HDL-c and LDL-c of the normal group compared with Zhiganfang-intervention group and Lipitor-intervention group (P<0.05). There is statistic significance of TG, TC, HDL-c and LDL-c of the model group compared with Zhiganfang-intervention group and Lipitor-intervention group (P< 0.05). There is no statistic significance between Zhiganfang-intervention group and Lipitor-intervention group(P>0.05). There is statistic significance of MDA of the model group compared with the other three groups(P <0.05). There is statistic significance between Zhiganfang-intervention group and Lipitor-intervention group(P<0.05). There is no statistic significance of SOD between model group and normal group(P>0.05). There is statistic significance of SOD of Zhiganfang-intervention group compared with model group and Lipitor-intervention group(P<0.05).There is statistic significance of ratio of SOD to MDA of the model group compared with the other three groups (P<0.05);and there is no statistic significance between Zhiganfang-intervention group and Lipitor-intervention group(P>0.05). There is statistic significance of expression of UCP2mRNA of the model group compared with the other three groups (P<0.01);and there is no statistic significance between Zhiganfang-intervention group and Lipitor-intervention group (P> 0.05) Conclusion:We successfully prepared rat model of NAFLD. There is function disorder of mitochondria, both liver local and systemic oxidative injury of the NAFLD rats. Zhiganfang can reduce the degree of liver steatosis, that might be partially dependent on the antioxidant capacity of liver, Zhiganfang prevents liver from oxidative injury, and thus delays non-alcoholic steatohepatitis progress. Function disorder of mitochondria is an important pathophysiologicals mechanism to promote the progress of NAFLD, and this therapy is a potential treatment for NAFLD.
