Objective:To investigate the effects of brain neurotransmitters and serotonergic system in mice loaded with restraint stress and the intervention of essence of chicken. Methods:(1) To establish different stress models of mice by taken blood in conscious conditions and restraint stressed 3 hours. To evaluate the effect of different types of stress on brain in mice by detected the concentrations of the histamine, dopamine (DA) and 5-hydroxytryptamine (5-HT) of plasma, cerebral cortex and midbrain. (2) To evaluate the relationship of the stress response and the central serotonergic system by observed the levels of 5-HT,5-hydroxyindoleacetic acid (5-HIAA) and tryptophan hydroxylase (TPH) activity of cerebral cortex and hippocampus of different restraint interval of mice. (3) Mice were stressed by acute restraint stress 18 hours, the step-through passive avoidance task was used to detect the learning and memory of mice, and then detect the levels of 5-HT,5-HIAA, tryptophan hydroxylase activity, the express of tryptophan hydroxylase-2 (TPH-2) and serotonin transporter (SERT) mRNA of cerebral cortex, hippocampus and plasma; and the intervention of essence of chicken.Results:(1) Taken blood in conscious conditions and restraint stressed 3 hours significantly increased histamine,5-HT, and DA level in different brain regions in mice (P<0.05 or P<0.01); the taken blood in conscious conditions and restraint stressed 3 hours significantly increased histamine level in stressed mice (P<0.01); however, the restraint stressed 3 hours group significantly decreased plasma levels of 5-HT (P<0.01). (2)After restraint stressed 4,8,12 hours of mice, the level of 5-HT,5HIAA and the tryptophan hydroxylase activity were significantly increased (P<0.05 or P<0.01); and after restraint stressed 18,24 hours of mice, the levels of 5-HT,5HIAA and the tryptophan hydroxylase activity were significantly decreased (P<0.05). (3) It were significantly decreased the ability of learning and memory, the levels of 5-HT,5-HIAA of plasma and brain in restraint stress mice (P<0.05 or P<0.01), and the TPH activity and the express of TPH-2 mRNA also were also significantly decreased (P<0.05 or P<0.01). However, oral administration the essence of chicken significantly increased the ability of learning and memory, the levels of 5-HT,5-HIAA, the TPH activity and the express of TPH-2 mRNA (P<0.05 or P<0.01).Conclusions:(1) Different types of stress have an impact on the central nervous system neurotransmitter, and the changes may be related to activate the central nervous system stress-related.(2) Restraint stress can affect the central serotonergic system of mice, thereby affecting the level of brain 5-HT and tryptophan hydroxylase activity. (3) The essence of chicken can increased the ability of learning and memory of impairment in restraint stress mice, and the effect may be activation of the central serotonergic system through enhanced the level of 5-HT, tryptophan hydroxylase activity, tryptophan hydroxylase-2 mRNA expression to achieve.
