OBJECTIVE1. To establish the analysis of the omethoate in biomaterial by a GC and a GC/MS.2. To develop omethoate intragastric administration death animal models, and observe the mainly vital signs of dogs after an intragastric administration with omethoate.3. To investigate the detection of omethoate in dead dogs to provide a scientific evidence for the forensic identification of the poisoning accident caused by omethoate.METHOD1. GC/MS and GC:The samples including blood and tissues, to which internal standard object (malathion) was added After precipitating protein with 0.5mL 10% trichloroacetic acid, Omethoate was extracted with dichloromethane and analyzed qualitatively and quantitatively by GC/MS and GC respectively.2. The detection of omethoate:8 dogs were given an intragastric administration of omethoate with a dose of 14mg·kg-1. As soon as the vital signs disappeared, the dogs were dissected and the specimens such as heart blood, heart, liver, spleen, lung, kidney, skeletal muscle, brain, bile, urine were sampled immediately, in which the concentration of omethoate was determined by a GC and a GC/MS.3. Statistics method:The data were collected by software SPSS 11.5 and expressed by x±s.RESULTS1. Symptoms:salivation, lacrimation, urinary and fecal incontinence, vomiting, pupillary miosis, spasm, fasciculation and dyspnoea occureed at 10-20 minites after an intragastric administration with omethoate. And all dogs from intragastric administration with omethoate died after 36.63±16.87minites. 2. The content of omethoate in dogs:the average content of omethoate from high to low:liver, heart, lung, brain, skeletal muscle, kidney, spleen, heart blood, urine, bile.CONCLUSION1. The GC/MS and GC method is more selective, more sensitive, simpler, more accurate, and can be applied not only to the clinical diagnosis of omethoate poisoning, but also to forensic identification of omethoate poisoning death.2. Besides the normal specimens stomach, stomach contents and heart-blood, we still collect other tissues such as liver, heart, lung and brain in order to be more full-scale qualitation and quantitative analysis.Then we get to conclusion combining with symptom and physical sign.
