Insulin resistance is an important pathophysiological basis of cardiovascular and endocrine diseases. Numerous studies in recent years have focused on the relationship between insulin resistance and coronary heart disease, hypertension, hyperglycemia, hyperlipidemia, and vascular inflammation, While research of myocardium change under insulin resistance is so rare. In this paper, with a simple approach to building high-fat-fed rat model of insulin resistance, and the use of ARB telmisartan to interfere with the early myocardial inflammation under insulin resistance.Materials and methods40 Male Wistar rats are randomly divided into 2 groups:control group and the high fat group. By feeding we construct rat model of insulin resistance. Diagnosed with insulin resistance "gold standard"-"clamp experiment" on two group rats to determine the presence of insulin resistance. Model high-fat group were divided into intervention group and non-intervention group, Intervention approach to drug intervention by gavage for 8 weeks. After 4 weeks and 8 weeks respectively, rats of three groups are randomly detected insulin resistance state and obtained the myocardial tissue and blood samples. Biochemical method to detect serum insulin and lipid levels, immunohistochemical method to detect of cardiac IL-6 levels.All data were analyzed with statistical software SPSS13.0, if data is continuous measurement, expressed byx±s, t test is used to compare the two groups.Result1. glucose infusion rate:high-fat diet for 8 weeks, the control group is significantly higher than high fat group (P<0.05); telmisartan intervention for 4 weeks : the intervention group is significantly higher than non-intervention group but was still lower than the control group (P<0.05); Telmisartan intervention for 8 weeks: the intervention group is significantly higher than non-intervention group but still significantly lower than the control group (P<0.05);2. Electron microscopy results:high-fat diet for 8 weeks myocardial fiber gaps are irregular and most expand, the sarcomere Z line is still clear, mitochondria between adjacent muscle fibers decrease, outer membrane damages, intervention after 4 weeks, micro-myocardial cells structure as a whole is better than that before intervention, significantly better than non-intervention group and intervention for 8 weeks the results are more clear.3. Inflammatory cytokines IL-6 expression:the normal tissue cells do not express, and not expressed in control myocardium. Expression of the intervention group was not obvious, the non-intervention group expresses strongly positive. There were significant differences between the colored rate of cytoplasm of IL-6 expressed in non-intervention group and the control group (P<0.05); there is no significant difference between intervention group and the control group, there is no statistical difference(P<0.05).conclusion1. IL-6 expression in rat cardiac under insulin resistance enhances;2. Addition to control of blood glucose and blood lipid lowering effect, ARB can improve insulin resistance;3. ARB may improve chronic inflammatory response underinsulin resistance.
