| Objective: In this experiment, the high-fat diet of OLETF rats, insulin resistance(IR) experimental animal model after the success of drug intervention with telmisartan.the purposes of this experiment 1.To determine the levels of expressions of Cav1.2in the atrial myocardium of rats with IR. To study the effect of AT1 R telmisartan on IR and the expression of Cav1.2; 3.To research the possible mechanisms of the telmisartan on the expression of atrial muscle protein Cav1.2 and improved IR.Methods: 16 LETO rats and 32 OLETF rats,under the condition of the standard animal feed and clean environment, adapt to feeding. In two months,the OLETF rats were fed with high-fat diet, in four months at the age of stop eating 12 hours,the plasma levels of FBG,FINS were measured,and HOMA-IR were evaluated,which to analysis whether IR model was successfully established. All the IR rats divided into 2 groups:16 rats in model group(M) to rats by high fat diet;16 telmisartan(T) rats fed with the high-fat diet combined with telmisartan(5mg/kg d) intervention, 16 LETO rats as normal control group(NC) fed with normal diet. According to the method of feeding rats after five months, rats used again to stop eating 12 hours after the blood samples were collected from the femoral vein, FINS detected by radioimmunoassay, enzyme method to detect FBG glucose, triglyceride detection using automatic biochemical analyzer TG, HDL-C, TC and LDL-C. Then use the spinal dislocation method executed by rats, quickly remove the experimental animal organs,using the Western blotting to detect the expression of Cav1.2 in atrium. Using SPSS20.0 statistical software, the results are calculated.Results: 1.Comparisons of three groups of experimental rats with FINS, FBG, HOMA-IR: compared with M group, FINS, FBG, HOMA-IR were significantly decreased in T group; compared with NC group, FINS, FBG, HOMA-IR value was significantly increased in group M,the difference was statistically significant(P< 0.05) 2.Comparison of experimental lipid metabolism in rats: a comparison between the three groups, T group and M group LDL-C, TG, TC values decreased significantly, the difference was statistically significant(P<0.05). But there was no significant difference in the HDL-C value, the difference was not statistically significant(P=0.384); group M, TLDL-C, TG, TC were significantly higher than that of group NC, HDL-C value was lower than that in NC group. 3.Comparisons of levels of Cav1.2 in atrium sinistrum tissue among every group: compared with T group and M group, Cav1.2 protein level in atrial tissue was significantly increased, compared with M group and NC group, Cav1.2 protein levels in atrial tissue were significantly decreased, the difference was statistically significant(P<0.05).Conclusion: 1.Successfully established by feeding high fat insulin resistant rats in serum OLETF FINS, FBG, HOMA-IR, TG, LDL-C and TC values were significantly higher, significantly lower HDL-C values. 2. After telmisartan, rat serum FBG, FINS, HOMA-IR, TC, TG and LDL-C levels were significantly lower, indicating that IR rats with telmisartan by improving glucose and lipid metabolism, improve insulin resistance. 3. In addition, atrial myocardium Cav1.2 is down-regulated in IR rats; After telmisartan intervention, the expression level of atrial myocardium Cav1.2 increase in IR rats. |
