| The liver is an organ of paramount importance that plays an essential role in the metabolism of protein, fat, sugar, vitamins, hormones, bile pigments and other substances. The liver is the important target of drugs, xenobiotics and oxidative toxicant. The liver injury is caused by various harmful factors, such as hepatitis virus, drugs, ischemia and so on.Liver injury is part of the process of liver pathology that is the starting link and common approach of occurrence and development of serious liver diseases. In addition, reduce the harmful factors of liver disease to protect the liver for improving the quality of life is essential.Object: Many epidemiological surveys and in vitro and in vivo experimental studies have demonstrated the protective effects of naturally antioxidants from injury by chemical poisons. The blueberry is rich in anthocyanins and has high antioxidant power.This study will investigate the effects of blueberry extract on acute chemical liver injury induced by Carbon tetrachloride using ICR mice as the acute chemical liver Injury exeperimental model.Method:According to"Norms on Health-care Food Examination and Technology Assessment"of China's Ministry of Health,ICR adult female mice were divided into 5 groups randomly by weight, each at least 10 including normal group, model group,grape seed anthocyanins group(30mg/kgBW), low blueberry extract group(30mg/kgBW) and high blueberry extract group(60mg/kgBW).During the experiment, the three test groups were intragastrically administered grape seed anthocyanins or blueberry extract every day for 30 days, and the normal group and the model group were administered the same Volume of distilled water .After 30 days of administration, all mice were fasted for 16 hours,then mice of all groups except the normal group were administered edible vegetable oil containing1% of Carbon tetrachloride at a dose of 5ml per kg of bodyweight, the animals were sacrificed 24h after CCl4 administration. Then Transaminases (alanine transaminase—ALT and aspartate—AST)were measured in serum. Extent of lipid peroxidation was measured in the liver homogenateby quantifying the malondialdehyde (MDA) which is expressed as MDAnmol/g.. Superoxidedismutase (SOD) activity was determined as the ability of the enzyme to inhibit cytochrome'c'oxidation and the enzyme activity was expressed as SODunits/g protein. liver index were assayed and liver pathological examination was also implemented.Data were expressed as mean±S.E M. The data was analyzed by an analysis of one-way ANOVA. Probability levels of <0.05 were considered significant.Result:There was no significant alteration in body weight and weight gain during the experiment among all groups.Compared with the normal group, the serum ALT and AST activity in the liver injury group, the positive control group, the low dose and high dose group of blueberry extract increased significantly;The serum AST and ALT activities in all test group were lower. than the liver injury group ,but there was no difference among the test groups.Compared with the liver injury model group, the liver malondialdehyde content was much lower in normal group , Compared with the liver injury model group ,the liver MDA content of the positive control group,the low dose and high dose animals blueberry extract significantly decreased ,but there was no difference among the test groups.The liver superoxide dismutase content of the liver injury model group, low dose and high dose of blueberry extract group decreased significantly compared with the normal group, There was no significant alteration in the liver superoxide dismutase content between the positive control group and the normal group .The liver superoxide dismutase content in all test group were significantly higher. than the liver injury group; the liver SOD activity of blueberry extract low-dose group animals was lower than the positive control group, the liver SOD activity of blueberry extract high-dose group animals and the positive control group was not significant; There was no significant alteration in the liver superoxide dismutase content between Low-dose and high dose animals blueberry extrac group.The liver index of the liver injury model group was higher than that of the nomal group ,Compared with the liver injury model group, the liver index among all the tested groups showed no significant difference ,and but had no significant difference from the indexs of the three test groups.Sections of liver stained by H&E were examined for necrotic cells, inflamed cells, neutrophil infiltration, cellular hypertrophy and fatty infiltration. in the livers of the model group, In rats administered CCl4, the hepatic injury was marked and widespread and included fatty changes, cellular hypertrophy, and necrotic foci. Compared with the normal group ,the scores about liver cells in histopathologic of the liver injury group, the positive control group, the low dose and high dose group of blueberry extract were significantly higher while the positive control group and blueberry extract high-dose group significantly attenuated CCl4-induced liver damage. The histopathologic scores of liver cells no significant difference among the tested group.Conclusion: In this research, grape seed anthocyanins and blueberry extract can reducde elevation of serum AST/ ALTand attenuated the pool of liver cell cytoplasm, hydropic degeneration of liver cells, liver cell injury and necrosis of liver cell pathology by CCl4-induced liver damage. improve the antioxidant enzyme activity, reduce lipid peroxide formation / acceleration Clear.In short, blueberry extract have protective effects against acute chemical liver injury induced by carbon tetrachloride. Blueberry extract(60mg/kg) to alleviate acute chemical liver injury results close to grape seed anthocyanins(30mg/kg). |
