Protective Effects And Mechanisms Of Shaodilan Tablet On Chemical Liver Injury

Objective To observe the protective effects of Chinese herbs complex prescriptions of Shaodilan ( SDL) tablet against the liver injury induced by carbon tetrachloride(CCL4)in rats,to explore its possible mechanisms and to investigate results of the acute toxic test in order to afford experimental evidence for clinical therapy.Methord We establish the model of chemical rats liver injury by hyodermic injection of 25% CCL4 and peanut oil liquor(5ml/kg) once on the first and the fifth day. 112 Wistar rats are randomly divided into seven groups,that is,normal group,sham liver injury group,liver injury group,biphenyl dicarboxylate( BPD )contral group,and large,middle,small dose SDL treatment group ( 5.2g / kg-d, 2.6g/kg-d, 1.3g/kg-d) .Each rat in the large ,middle,small dose SDL group was poured into the stomach once a day for seven days.To measure the activity of alanine transaminase(ALT X superoxide dismutase ( SOD X mialondialdehyde (MDA X nitric oxide (NO) in serum ;To measure the expression of Bcl-2 and Bax in liver cells .We also observed the changes of liver weight index and liver histopathology. The experiment research of the acute toxic test of SDL tablet on rats to observe the LD50 or maximum tolerance dose(MTD) .Results The above mentioned items in liver injury group are significant difference with that in normal group and sham liver injury group (P<0.05 ) .It indicates that liver is seriously damaged during making model,while only hypodemic injection peanut oil conversely .SDL tablet can significantly decrease the serum level of ALI\ MDA. NO and raise the serum level of SOD.And the action of decreasing the serum level of ALT. MDANO and raising the serum level of SOD in large dose SDL group is significantly superior to BPD group (P<0.05 ) . In large, middle, small dose of SDL groups,the expression of liver cells Bcl-2 decrease in proper order, while Bax increase conversely,and the ratio of Bcl-2 and Bax decline.Liver weight index is going up in large,middle,small dose SDL in proper order.The dose dependent ralation among the SDL groups is seen.Histopathologic examination shows that SDL can relieve liver injury in rats induced by CCL4. LD50 is not observed because of medical density and volume capcity of rat's stomach that could make rats to be death are so limited.MTD is equal to 206.63 times of clinic dosage.Conclusions These results indicate that SDL tablet can stop the oxidation of the lipid,lower the transaminase enayones and NO,increase the ratio of Bcl-2 and Bax.This experiment shows that the SDL have effects in protecting and curing liver injury by chmicals,and has no sideeffects.
