Paraquat (PQ) is a high-efficiency herbicide widely used at present. Because its toxicological effects and pathogenic mechanism have not been completely known by now, there have been no special-effect medicine in the clinical treatment and the death rate by poisoning has been very high. Many patients die of ARDS caused by the acute lung injury at the early time and die of respiratory failure caused by the pulmonary interstitial fibrosis at the late time. In recent years, the cytokine in the research on the PQ caused acute lung damage has been universally approved. The relationship between TGF-β1 and tissue organ fibrosis is one of the most important accretion induction factors. Therefore, the realization of the treatment of the lung fibrosis via the repression of the expression of TGF-β1 has become the hot spot under discussion in the medical circle by now.The experiment is mainly made to observe the lung tissue by the chemical method of taking the lung HE chromosome, Masson chromosome and immune tissue in the 1st, 3rd, 7th, 14th, 21st and 28th days, respectively after making the rat PQ poisoning model via the mouth, measure the TGF-β1 content in the tissue and discuss the pathogenic mechanism and the curing effects of Xuebijing injection, ulinastatin and combined application of the two medicines on the PQ poisoning. At the same time, the HE chromosome of the liver and kidney of the rat is made to study the damages to the liver and kidney by the PQ poisoning and the curing effects of Xuebijing injection, ulinastatin and combined application of the two medicines on the PQ poisoning.In the experiment, 150 healthy Wistar rats for the experiment are chosen and provided by the Animal Center of Jilin University, without any limit on the male and female, with the weight of (250+20)g. The rats are divided into Group A: normal contrast group (n=30): 1ml biological salt water poured to the stomachs; Group B: PQ infected group (n=30): 40mg/kg PQ liquid (diluted to 1ml) poured to the stomachs to make the PQ poisoning model and pour the equal dosage of biological salt water to the belly everyday; Group C: Xuebijing injection treatment group (n=30): 40mg/kg PQ liquid (diluted to 1ml) poured to the stomachs and 10ml/kg Xuebijing injection injected to the belly twice everyday; Group D: Ulinastatin treatment group (n=30): 40mg/kg PQ liquid (diluted to 1ml) poured to the stomachs and 20000 iu/kg Ulinastatin injected to the belly twice everyday; Group E: Xuebijing injection+ Ulinastatin treatment group (n=30): 40mg/kg PQ liquid (diluted to 1ml) poured to the stomachs and 10ml/kg Xuebijing injection+ 20,000 iu/kg Ulinastatin injected to the belly twice everyday. Kill the rats in the 1st, 3rd, 7th, 14th, 21st and 28th days, respectively, take the HE chromosomes of the middle tissue of the right lung, liver and right kidney. The Masson chromosome and TGF-β1 immune chemical chromosome are carried out for the lung tissue and the grey value of TGF-β1 is measured for the statistical analysis.1. Observe the lung tissue of the PQ poisoned rat via the HE chromosome and Masson chromosome and it is found that the rat is obviously represented in the inflammation at the early time of the PQ poisoning, but there exist the different extents of fibrosis and it is mainly represented as the lung fibrosis at the late period of poisoning. With the TGF-β1 immune chemical chromosome and measurement of the grey value, it is found that the rat has the TGF-β1 content raised after it is poisoned. Xuebijing injection and Ulinastatin can obviously repress the expression and lung fibrosis of the PQ poisoned rat and the combined use of Xuebijing injection and Ulinastatin can achieve the best effects.2. It is found from the HE chromosome of the PQ poisoned rat that the denaturalization, necrosis and self-limiting of the liver cells occur after the rat is poisoned.The injury is occurd on the 3~7th day and it is recovered on the 28th day. Xuebijing injection and Ulinastatin can obviously reduce the degree of pathological changes of the liver of the PQ poisoned rat and the combined use of Xuebijing injection and Ulinastatin can achieve the better effects than the separate use.3. The pathological changes of the kidney of the PQ poisoned rat from the HE chromosome are mainly the damages to the renal tubule and all the tissues have the pathological changes. Only the trend of all the groups is consistent, that is, it is serious on the 1st day, the pathological changes are similar on 3rd~21st days, it becomes minor on the 28th day and no obvious difference between all the groups is observed.Although the experiment can preliminarily prove that Xuebijing injection and Ulinastatin have the good curing effects on the PQ poisoning, the study is not comprehensive due to the limits of some factors such as time and conditions, etc. and the technical means are not imperfect; therefore, the deep-going and objective research and argumentation shall be further made to prove the curing effects of Xuebijing injection and ulinastatin on the PQ poisoning.
