Objective: Microleakage is the main failure reasons of root canal treatment,therefore requires fully sealing between the filling materials and the root canals,this is a challenge for gutta-percha. Resilon/Epiphany Self-Etch is a new resin-base filling material, there is a chemical bond between the new material and the root canal wall, so it can reduce the microleakage . The purpose of this study was to compare the apical sealing ability of the two obturation Materials: Resilon / Epiphany Self-Etch and Gutta-percha / AH plus through the methods of dye leakage, serial sections and demineralization to choose a better root canal filling material than the gutta-percha and provide a theoretical basis for the clinical.Materials and methods: 36 recently extracted mandibular premolar roots were studied, 6 for the positive and negative groups, 30 for the experimental groups (14 for the Resilon/Epiphany SE groups, 16 for the Gutta-percha / AH plus groups).The roots was prepared to the same standards, then filling the root canals respectively with Resilon/Epiphany SE and Gutta-percha /AH plus. A X-ray film will be taked from the teeth of the buccolingual and mesiodistal after filling root canals . And then evaluate the X-ray film quality of the root canal filling materials. Then put the tooth into the India ink to stain, use hard tissue slice machine to make the tooth become some sharp slices. Use the microscope to observe the horizontal cross-section and record the stained of each section .The dye leakage will be observed under the scanning electron microscope to evaluate the sealing and bonding of the two materials, then the slice was demineralizate with nitric acid, scanning electron microscope was use to evaluate the bonding manner between the teeth and the two filling materials . Comparative the difference of the apical sealing ability of the two obturation materials .Results: 1. The buccolingual x-ray of the two kinds of filling materials'quality was higher than the mesiodistal's (P<0.05). There was no statistical significance between the two kinds of filling materials of the buccolingual x-ray quality (P>0.05). The mesiodistal x-ray quality of the Resilon/Epiphany SE was higher than the Gutta-percha /AH plus (P<0.05) .2. The sealer of the Resilon/Epiphany SE was bonding to the resin filling materials and the root canal dentin ,and become a structure name monoblock.When compare to the Gutta-percha /AH plus groups ,the Resilon/Epiphany SE groups have lower apical leakage (P<0.05) . But the Gutta-percha/AH plus groups display a gap between the filling materials and the root dentins ,and have higher apical leakage.Conclusion: 1. The results of this study showed that the Resilon/Epiphany SE filling system has good radiopacity that help us to observe its filling quality after root canal therapy. 2. The results of this study showed that the sealer of the Resilon/Epiphany SE was bonding to the resin filling materials and the root canal dentin ,and become a structure name monoblock.,and the Resilon/Epiphany SE filling system has better apical sealing ability than the traditional filling material of Gutta-percha/AH plus.3. This study only evaluate the micro-leakage of the Resilon / Epiphany SE filling system .Further investigation should be conducted to evaluate other properties of the new filling system to determine if the new obturation material could replace gutta-percha as a root canal filling material.
