Objective:To approach the inhibition of disintegrin (Kistrin) in proliferation of lens epithelial cells with rabbits'posterior capsule opacification(PCO) modles.Methods: 1,Thirty-six New Zealand rabbits eyes (each eye in each rabbit) were performed extracapsular lens extraction surgery (ECLE) then randomly divided into 4 groups.Group A was the control group while groups B,C and D were the experimental groups. Each group contained nine rabbits′eyes. After the extracapsular lens extraction, 0.2ml of Ringers solution and 0.2ml of three different concentration of Kistrin(40μg.L-1,80μg.L-1,160μg.L-1)were injected into the sugery eyes'anterior chamber of rabbits in groups A,B,C and D, respectively. After 14 days of operations, the eyeballs were removed and the proliferating cell nuclear antigen (PCNA)expression were detected by immunohistochemistry and the proliferative indexes (PI) were calculated (PI=PCNA positive cells/50×100%). In order to evaluate the toxicity of the most effective concentration of Kistrin on cornea and retina, HE staining and transmission electron microscopy were carried out .2,Twelve New Zealand rabbits (12 eyes) were performed extracapsular lens extraction surgery (ECLE) and randomly divided into 4 groups.The control group:A group and experimental groups B,C and D, Each group contained three rabbits′eyes. After the extracapsular lens extraction.we inject 0.2ml of Ringers solution and 0.2ml of three different concentration of Kistrin(40μg.L-1,80μg.L-1,160μg.L-1)were injected into the sugery eyes'anterior chamber of rabbits in groups A,B,C and D, respectively . After 3th months of operations, the eyeballs were removed and the proliferating cell nuclear antigen (PCNA) expression were detected by immunohistochemistry and the proliferative indexes (PI) were calculated (PI=PCNA positive cells/50×100%).Results:1,①PI index of Goup A,B,C and D were 0.32±0.02,0.175±0.03, 0.065±0.02, 0.19±0.05, respectively. 14 days after operation, the incidences of PCO were significantly decreased in the experimental groups compared with group A (P<0.01); compared with Group B and Group D, the PI index of Group C's was reduced significantly (P<0.01), while Group B wasn't found different from Group D (P>0.05.)②The PI indexes of equatorial areas in Group A,B,C and D were 0.39±0.06, 0.23±0.04,0.11±0.03,0.24±0.08, respectively, while the PI of central areas in those were 0.25±0.05,0.12±0.03,0.06±0.02,0.14±0.04, respectively.The PCNA positive expressions of the equatorial areas were higher than those in the central areas (P<0.01).③No difference in the corneas and the retinas were found in Group C when compared with Group A by using HE staining and Transmission electron microscopy.2,①PI index of Goup A,B,C and D were 1±0.000,0.94±0.004,0.02±0.001, 0.063±0.007. respectively.3th months after operation.②Compared with Group A, the PI index of Group C and Group D were reduced significantly (P<0.05).③compared with Group B and Group D, the PI index of Group C's was reduced significantly (P<0.05), Conclusion:1,Kistrin(40μg.L-1,80μg.L-1,160μg.L-1)can effectivily suppression the PCO formation in early stage. Kistrin(80μg.L-1,160μg.L-1)can inhibit the expression of PCNA and reduced the formation of PCO 3th months after operation.2,The equatorial LECs was stronger than the central part at the early formation of PCO.3,80μg.L-1Kistrin is a candidate concentration for the Kistrin in studying its inhibition of PCO formation., this concentration drugs can be used for the next experiment.
